摘要:
目的
对武当山区重楼属植物进行种内分子鉴定研究,以DNA条形码技术解决重楼因种内形态差异小而难以鉴别的问题,规范重楼的正品来源,确保本地药材的质量以及临床用药的有效性。
方法
对ITS(internal transcribed spacers)序列进行克隆测序,以核基因ITS2片段作为DNA条形码,对研究材料进行PCR(polymerase chain reaction)扩增并双向测序,所得序列经CodonCode Aligner拼接后,用软件MEGA6.0(Molecular Evolutionary Genetics Analysis 6.0)进行相关数据分析,并构建NJ邻接树(neighbor-joining tree),利用Keller等建立的ITS2数据库及其网站预测ITS2的二级结构。
结果
经BLAST鉴定,采集的武当山区重楼样本来源准确;武当山区重楼属植物ITS2序列长度均为232 bp,与七叶一枝花的基因序列相比,球药隔重楼有16个变异位点,华重楼只有1个变异位点;武当山区重楼的ITS2序列种内K2P(Kimura-2-parameter)距离为0~0.073 6,种内平均K2P距离为0.027 2;由所构建的系统聚类树可以看出,本地区重楼主要分为七叶一枝花和球药隔重楼2大类,华重楼为七叶一枝花的变种,宽叶重楼、狭叶重楼与七叶一枝花为一支,不支持宽叶重楼、狭叶重楼成为独立的变种;比较ITS2二级结构发现,武当山区重楼属植物螺旋区的茎环数目、大小、位置有明显差异。
结论
DNA条形码为重楼属药用植物鉴别开辟一条新途径,不论是相似性搜索法、最近距离法还是NJ系统发育树等方法,其分析结果均充分表明,ITS2序列可将不同品种重楼药材很好地区分开;七叶一枝花、狭叶重楼、宽叶重楼的ITS2基因232个碱基序列完全相同,MEGA 6.0的分析结果也表明宽叶重楼、狭叶重楼与七叶一枝花属于一支,不支持宽叶重楼、狭叶重楼成为独立的变种,建议作为七叶一枝花的变型处理;ITS2作为DNA条形码序列能够有效地区别重楼属植物,在中药材的鉴定中具有重要的应用前景,在实现规范化种植规程中可用于种质种源的正确鉴定。

Abstract:
Objective
DNA barcoding technology, a molecular identification method, is applied to Paris polyphylla collected from Wudang Mountain area in order to solve the differential problem caused by small morphology difference, make authentic origins canonical, and guarantee the quality and clinical curative effect of this medical material in our area.
Methods
In this study, the second internal transcribed spacers (ITS2) of ribosomal DNA were amplified and sequenced. Sequence assembly and consensus sequence generation were performed using the CodonCode Aligner. To perform a phylogenetic study, the genetic distances were computed using molecular evolutionary genetics analysis 6.0 (MEGA 6.0) in accordance with the Kimura-2-parameter (K2P) model. The phylogenetic tree was constructed using the neighbor-joining (NJ) and Kimura-2-parameter methods. The ITS2 secondary structures were predicted using ITS2 database and website which was found by Keller.
Results
Identified by BLAST, P. polyphylla collected in Wudang mountain area was proved to be precise. The results showed that the ITS2 sequence length was 232 bp. It was indicated that their K2P distance were 0-0.073 6 and the average was 0.027 2. Compared with the gene sequence of P. polyphylla, there were 16 variation loci in P. fargesii and only one variation locus in P. polyphylla var. chinensis. As seen from the NJ tree, P. polyphylla in this area was mainly divided into two categories, P. polyphylla and P. fargesii. Obviously, P. polyphylla var. chinensis was a variation of P. polyphylla. Meanwhile, it was not tenable that P. polyphylla var. stenophylla and P. polyphylla var. latifolia were the variations of P. polyphylla because they belonged to a branch. To compare the ITS2 secondary structure of P. polyphylla, we noticed the differences in the amount, size, and position of loop of helix zone.
Conclusion
DNA barcoding technology opens up a new way for the identification of P. polyphylla. Regardless of what methods were used, such as similarity search, nearest distance, and NJ tree method, the analysis results have fully demonstrated that ITS2 sequences could be used to correctly identify the different varieties of P. polyphylla. It is identical in 232 base sequence of ITS2 to P. polyphylla var. stenophylla, P. polyphylla var. latifolia, and P. polyphylla. The results computed by Mega 6.0 also show that they belong to a branch. It is more appropriate to say P. polyphylla var. stenophylla and P. polyphylla var. latifolia are variants of P. polyphylla. Hence, as a DNA barcode sequence, ITS2 can be applied to intraspecific discrimination of P. polyphylla. Furthermore, the application of ITS2 in the identification of traditional Chinese medicine has an important prospect. And it can be used to correct identification of germplasm resources in GAP procedures.

重楼是百合科重楼属植物的统称,最早以蚤休之名始载于《神农本草经》,列为下品[1]。重楼主要分布于欧洲和亚洲温带、亚热带地区,约有10个品种,我国分布有7种和8变种[2],《中国药典》2015年版收载的正品重楼为百合科植物云南重楼Paris polyphylla Smith var. yunnanensis (Franch.) Hand.-Mazz.或七叶一枝花P. polyphylla Smith var. chinensis (Franch.) Hara的干燥根茎[3]。重楼具有抗肿瘤、镇静镇痛、止咳平喘、免疫调节、止血等作用[4-5],是云南白药、季德胜蛇药的主要原料,需求量很大,除《中国药典》2015年版收载的品种外,其余多数品种均在市面上流通[6-7]。为了更好地规范重楼的正品来源,确保本地药材的质量以及临床用药的有效性,有必要对重楼正品来源与本地混用品种进行种内鉴别研究。
几种分子方法已应用于识别药用植物[8]。DNA条形码技术通过比较DNA条形码,从已知物种的参考序列库中查询未知样本进行真核生物的识别和鉴定[9-11]。该方法简便高效,不受样品的形态性状以及研究者的专业水平限制,适用于来源于生物体任何发育阶段部分组织或器官的中药材的真伪鉴别[12]。最理想的DNA条形码区域是在一个小片段出现所有物种的一个主要分类,即相同物种的所有成员拥有不变的核苷酸序列,但有足够的变异物种之间的区别[13]。朱英杰等[14]对重楼属11个物种17份样品的psbA-trnH、rpoB、rpoC1、rbcL、matK和核ITS2序列进行PCR扩增和测序,比较分析结果显示,ITS2序列在所研究的重楼属药用植物中的扩增和测序效率均为100%,对于ITS2序列扩大至29个物种67份样品依然具有100%的鉴定成功率,显示ITS2序列能够准确鉴定重楼属药用植物,可以作为潜在的重楼属药用植物通用条形码序列。本实验选择较易获得且方便处理的重楼属植物叶片作为实验对象,利用ITS2序列对采自武当山区重楼原植物进行DNA条形码鉴别研究,为该药材的准确鉴别提供分子证据。
1 仪器与试药
1.1 仪器与试剂 球磨仪(MM400,德国莱驰),冰箱(BCD-241WDBB,中国海尔),水浴锅(HHS-4S,上海虞龙),小型高速离心机(5424R,德国艾本德有限公司),PCR仪(PTC-200,美国伯乐),电泳仪(JY1000C,北京君意)。无水乙醇、异丙醇、氯仿、β-巯基乙醇等试剂均为分析纯。
1.2 药材 重楼样品采自武当山区和神农架林区,主要包括湖北省十堰市辖区内张湾区、茅箭区、郧阳区、竹山县、竹溪县、房县、郧西县、丹江口市、神农架林区,采集的植物叶片用吸水纸包好放入装有硅胶的自封袋中进行干燥保存,备用。材料均经湖北医药学院生药学教研室主任陈吉炎教授鉴定,鉴定结果见表 1。常见混伪品的信息均从GenBank中获得,见表 1。

2 方法
2.1 DNA提取 取植物叶片约30 mg,置2.0 mL的Ep管中,在球磨仪中充分研磨成粉末。加入600 μL在65 ℃水浴锅里预热的Buffer PCR和12 μL的β-巯基乙醇,于65 ℃水浴25 min,间或混匀。水浴后加入20 μL的RNase A(10 mg/mL)室温放置2~5 min后,加入等体积的氯仿,充分混匀,12 000 r/min离心5 min。吸取上层水相至1.5 mL离心管中,加入等体积Buffer BD,颠倒混匀3~5次,再加等体积的无水乙醇,充分混匀后用移液器将其全部加入到吸附柱中,室温静置2 min,10 000 r/min离心1 min,倒掉收集管中废液。将吸附柱放回收集管中,依次加入500 μL PW Solution、500 μL Wash Solution,同法处理倒掉收集管中废液后,再将吸附柱放回收集管中,12 000 r/min离心2 min,打开吸附柱盖子于室温放置数分钟,彻底晾干吸附材料中残留的Wash Solution。将吸附柱放入一个新的1.5 mL的离心管中,在吸附膜中央加入50 μL TE Buffer,静置3 min,12 000 r/min离心2 min,得到的DNA溶液置于-20 ℃保存,备用。
2.2 PCR扩增及测序 采用核基因组的rDNA-ITS区的引物2F及3R(引物序列为2F:5’-ATGCGATACTTGGTGTGAAT-3’位于5.8 S上;3R:5’-GACGCTTCTCCAGACTA-CAAT-3’位于28 S上)进行序列扩增,引物由上海生工生物工程股份有限公司合成。
扩增反应体系体积为25 μL,包括2×Taq PCR Master Mix 12.5 μL,正反向引物各1 μL(2.5 μmol/L),模板DNA为2 μL,加灭菌超纯水至25 μL。PCR反应程序为95 ℃预变性5 min;94 ℃变性30 s,56 ℃退火30 s,72 ℃延伸45 s,40个循环;72 ℃后延伸10 min,4 ℃保存。取扩增产物5 μL与2 mL稀释后的SYBR Green I染料混合后加入到1.0%琼脂糖凝胶的点样孔中,电泳1 h后将凝胶置于紫外灯下观察,将有明显条带的扩增产物置于4 ℃冰箱中暂时保存,送检。
PCR产物送交上海生工生物工程股份有限公司测定,分别以2F、3R引物为测序引物,进行DNA序列的双向测定,以保证测定序列的准确性。
2.3 数据处理 测序峰图利用CodonCode Aligner 4.2.4校对拼接,去除引物区获得ITS2间隔区序列。将测定的基因序列提交到GenBank数据库,在数据库中的登录号为KR152196-KR152209。首先,运用NCBI Blast工具完成物种鉴定。然后,将所有序列用软件MEGA6.0进行多序列对比、变异位点分析并计算K2P(Kimura-2-parameter)遗传距离,用邻接(NJ)法构建系统聚类树,利用Bootstrap(1 000次重复)检验各分支的支持率。最后,在ITS2数据库中预测武当山区重楼ITS2的二级结构。
3 结果与分析
3.1 相似性搜索法鉴定结果 进入DNA条形码鉴定系统网站(http://blast.ncbi.nlm.nih.gov/Blast.cgi),对实验药材的ITS2序列使用BLAST方法进行鉴定和序列验证,结果显示实验序列查询到所有序列均属于重楼属,此鉴定结果说明实验药材样本来源准确。
3.2 一级序列分析 采自武当山地区和神农架林区的重楼样本ITS2序列长度均为232 bp,14个重楼属植物样品可以划分为3个组群:样品1、4、13号碱基序列相同,原植物鉴定为球药隔重楼;2、3、5、6、7、8、9、10、14号碱基序列相同,其中2、3、7、9、10号鉴定为七叶一枝花,5、6、8号鉴定为宽叶重楼,14号鉴定为狭叶重楼;11、12号碱基序列相同,相对于七叶一枝花的序列在163位点有T-C变异,鉴别为七叶一枝花的变种华重楼。ITS2基因序列的结果不支持宽叶重楼、狭叶重楼成为独立的变种,建议作为七叶一枝花的变型处理。以药典正品重楼七叶一枝花的序列为标准,各样品ITS2序列变异位点如表 2所示。

3.2.1 变异位点 以《中国药典》2015年版正品重楼七叶一枝花的序列为标准,球药隔重楼有16个变异位点,显示球药隔重楼与七叶一枝花确为2个不同种;而华重楼只有在163位的1个变异位点,这一点与华重楼为七叶一枝花的变种相符。
3.2.2 遗传距离 对ITS2序列进行种内K2P距离分析可知,重楼的ITS2序列种内K2P距离为0~0.073 6,种内平均K2P距离为0.027 2。宽叶重楼、狭叶重楼与七叶一枝花的K2P为0,又同为重楼属植物,确定为同一品种,印证了序列比较的结果。球药隔重楼与七叶一枝花的K2P为0.073 6,遗传距离最远,与变异位点分析结果相符。球药隔重楼与华重楼的K2P为0.068 7,遗传距离较七叶一枝花近。七叶一枝花与华重楼的K2P为0.004 3,说明华重楼为七叶一枝花的变种这一结论成立。
3.2.3 聚类分析 对武当山区的14种重楼及其常见混伪品进行聚类分析,NJ树见图 1。结果显示14种重楼聚为一支,拳参、延龄草和蜘蛛抱蛋单独聚为一支,说明采集样本来源准确,且本地区重楼主要分为七叶一枝花和球药隔重楼2大类,华重楼为七叶一枝花的变种,宽叶重楼、狭叶重楼与七叶一枝花为一支,不支持宽叶重楼、狭叶重楼成为独立的变种,这与序列比较分析结果一致。
3.3 二级结构分析 在ITS2数据库(http://its2.bioapps.biozentrum.uniwuerzburg.de.)[15]输入测得序列预测武当山区重楼ITS2二级结构,见图 2。由图 2可以看出,重楼属植物的二级结构均为一个中心环(主环)及4个螺旋区构成,每个螺旋上又有大大小小、或多或少的茎环结构。通过比较七叶一枝花与华重楼的ITS2二级结构发现,二者的差异较小,主要鉴别特征区为Ⅲ的C区,七叶一枝花比华重楼多一个茎环;通过比较七叶一枝花与球药隔重楼的ITS2二级结构发现,二者的差异较大,Ⅰ的A区、Ⅲ的B区和C区均可有效鉴别这两种重楼,七叶一枝花的A区比球药隔重楼多2个茎环,C区比球药隔重楼多1个茎环;B区七叶一枝花为1个单独的茎环,而球药隔重楼为一大一小相连的茎环。七叶一枝花与华重楼的二级结构只在C区有一个茎环的差别。因此,依据ITS2二级结构可以直观地将重楼属植物区分。
4 讨论
4.1 DNA条形码为重楼属药用植物鉴别开辟一条新途径 传统重楼鉴别有原植物鉴别、显微鉴别和理化鉴别。这些鉴别方法都依赖于鉴别人员的经验和操作技术,辨别药用植物的种属有很强的主观因素。然而,重楼属植物种内形态差异小,鉴别性状存在交叉或重叠[16],难以准确鉴别。DNA条形码这一新型的科学手段为重楼属药用植物鉴别开辟了一条新路径。不论是相似性搜索法、最近距离法还是NJ系统发育树等方法,其分析结果均充分表明,ITS2序列可将不同品种重楼药材很好地区分开来。因此,基于ITS2序列建立的DNA条形码鉴定技术,可以为市场上重楼药材的混用现象提供有效的技术方法和解决途径,为保障中医临床用药安全以及用药准确性提供技术支持。
4.2 ITS2作为DNA条形码序列能有效区别重楼属植物 重楼是经济价值较高、市场缺口较大的名贵中药[17],由于重楼的生长环境要求严格,生长缓慢、自然生长周期长、繁殖系数低,长期大量采挖使野生资源日益减少[18]。随着药材用量的不断增加,野生资源渐趋枯竭,重楼价格一路飙升至2 000元/kg。由于价格的刺激,难免有混伪品进入市场,为了更好地规范重楼的正品来源,确保本地药材的质量以及临床用药的有效性,本研究采集了武当山区各地的重楼,利用目前被广泛认可的DNA条形码ITS2序列片段,对重楼属植物进行种内鉴别,发现重楼的ITS2条形码序列长度均为232 bp,重楼的ITS2序列种内K2P距离为0~0.073 6,种内平均K2P距离为0.027 2,在基于邻接法构建的系统聚类树中,重楼属植物能明显区分开,ITS2二级结构又进一步显示了重楼属植物所具有的明显差异,因此,利用ITS2序列作为DNA条形码,可以有效地鉴定重楼。
4.3 实验结果不支持宽叶重楼、狭叶重楼成为独立的变种 马剑等[19]建议matK基因碱基差异为“0”的重楼作为变型处理较为恰当。从本研究结果看,七叶一枝花、狭叶重楼、宽叶重楼的ITS2基因232个碱基序列完全相同,MEGA 6.0的分析结果也表明宽叶重楼、狭叶重楼与七叶一枝花属于一支,不支持宽叶重楼、狭叶重楼成为独立的变种,建议作为七叶一枝花的变型处理。对于这种基因型相同,表型却有差异的现象,可能是由于本地环境因素的影响造成营养器官的变异,这也恰恰说明了DNA条形码应用于药用植物鉴别的客观性和准确性,减少主观因素造成的影响,从而规范重楼的正品来源,确保本地重楼药材的质量以及临床用药的有效性。
