[摘要]
目的通过不同截留相对分子质量的超滤膜对地黄多糖分离纯化,再利用纳滤膜对超滤液进行浓缩纯化。方法采用不同截留相对分子质量超滤再纳滤的工艺流程。结果两次超滤得到低聚糖,最佳操作条件为:料液质量浓度为13~132mg/mL,操作压力为0.25~0.275MPa,温度20~40℃。然后采用纳滤膜对超滤液进行浓缩纯化,适宜工艺条件为:料温为20~40℃,操作压力0.59~0.79MPa,浓缩倍数可达3倍。应用超滤-纳滤技术,低聚糖得率为46.63%,质量分数达到93.3%,凝胶过滤法验证低聚糖制品的相对分子质量低于6000。结论该工艺能有效分离纯化地黄低聚糖,简单可行,操作简便。

ObjectiveOligo saccharides of Rehmann iaglutinosa were separated and purified by different molecular weight cut-off ultrafiltration (UF) membranes, and then ultraf iltrate was concen trated and purified by nanofiltration (N F) membrane.MethodsDifferent molecular weight cut-off U Fun it first and then N Funit were used in process. ResultsOligo saccharides were separated by twice UF, the optimum separation conditions: the concen tration of feed solution was 13-132 mgöm L , the operation pressure was 0125-01275M Pa, and the temperature was 20-40 ℃1 Then ultrafilt rate was concen trated and purified by NF membrane, the optimum separation conditions: the operation pressure was 0159-0179 MPa, the temperature was 20-40 ℃, and the concentration multip legot to three.Under this condition,the total extraction rate of oligo saccharide products was 46163% and the purity was above 9313% ,the molecular weights of oligo saccharide product sdetermined by gelfilt ration were less than 6000.ConclusionThe technology is not only simple and feasible but also easy to separate and purify the oligo saccharides of Rglutinosa effectively.
