[摘要]
螺旋藻藻粉经热水抽提,乙醇沉淀,Sevag法去蛋白质,十六烷基三甲基溴化铵(CTAB)沉淀得酸性杂多糖,再经DEAE-纤维素柱层析分级纯化两次得多糖PSP1和PSP2。PSP1和PSP2经HPLC的Carbohy drate Analysis柱层析为单一对称峰,证明为均一多糖。纸层析和硫酸-咔唑反应分析表明:PSP1主要由D-半乳糖、D-甘露糖、葡萄糖醛酸及D-葡萄糖四种残基组成,比例为2.1∶3.0∶1.9∶2.8;PSP2主要由D-甘露糖和葡萄糖醛酸两种残基组成,比例为6.2∶3.8。由粘度法测得其分子量分别为12400和16800。IR、1HNMR和13CNMR证明:SP11主要含α型糖苷键及部分β型糖苷键,PSP2主要含α型糖苷键。

Crude polysaccharides were prepared by ethanol precipitation of the hot water extract of powdered Spirulina platensis and deproteinized by Sevag method. Complex acidic heteropolysaccharides of S. platensis (PSP1) and (PSP2) were then obtained by further treatment with cetyltrimethylammonium bromide (CTAB) and chromatographed on DEAE-cellulose column. Their homogeneity was proved by HPLC carbohydrate analysis. Their mean molecular weights were estimated to be 12 400 and 16 800, respectively. According to paper partition chromatography (PPC) and Dische analysis, PSP1 proved to be composed of D-galactose, D-mannose, glucuronic acid and D-glucose in a molar ratio of about 2.1∶3.0∶1.9∶2.8 and PSP2 was composed of D-mannose and glucuronic acid in a molar ratio of about 6.2∶3.8. IR, 1HNMR and 13CNMR indicated that PSP1 had α and β configurations, and PSP2 had α configuration.
