[摘要]
目的 建立HPLC测定壮药羊开口中3,3',4'-三甲氧基鞣花酸-4-O-β-D-吡喃葡萄糖苷的含量方法。方法 采用Welch Materials Ultimate AQ C18反相色谱柱(250 mm×4.6 mm,5 μm);流动相为乙腈-0.2%磷酸水溶液,采用梯度洗脱,体积流量为1.0 mL/min,检测波长为246 nm,柱温为35℃。结果 3,3',4'-三甲氧基鞣花酸-4-O-β-D-吡喃葡萄糖苷进样量在0.02~0.60μg内与峰面积线性关系良好(r=1.000 0);平均回收率为105.58%,RSD为2.27%。结论 该方法简便、重复性好、准确度高,可用于壮药羊开口的质量控制。

Objective To establish a HPLC method for the determination of 4-O-β-D-glucopyranosyl-3,3',4'-tri-O-methylellagic acid in Zhuang Medicine Radix Melastomatis normalis. Methods A Welch Materials Ultimate AQ C18 reverse phase column(250 mm×4.6 mm, 5 μm)was used. The mobile phase consisted of acetonitrile-water containing 0.2% phosphoric acid solution with gradient elution at a flow rate of 1.0 mL/min. The detection wavelength was set at 246 nm and the temperature of column was 35 ℃. Results The linear relationship between 4-O-β-D-glucopyranosyl-3,3',4'-tri-O-methylellagic acid injection amount and peak area was good in the range of 0.02—0.60 μg (r=1.000 0). The average recovery was 105.58% with RSD of 2.27%. Conclusion The method is simple, reproducible and accurate, and can be used for the quality control of Zhuang Medicine Radix Melastomatis normalis.
