[摘要]
目的 研究长春花Catharanthus roseus全草的化学成分及其抗肿瘤活性。方法 运用硅胶柱色谱、ODS柱色谱及HPLC等手段分离纯化化合物,并通过NMR和HR-ESI-MS等谱学技术进行结构鉴定;同时利用CCK-8法评估其体外抗肿瘤活性。结果 从长春花95%乙醇提取物中得到6个生物碱,分别鉴定为长春花碱A(1a)、2,3,3a,5-四氢-1H-苯并[d]吡咯并[2,1-b][1,3]恶嗪-1-酮(1b)、β-咔啉-1-羧酸(2)、1-乙酰基-β-咔啉(3)、β-咔啉生物碱(4)、2-(2-甲基-3-烯-2-基)-1H-吲哚-3-甲醛(5)。结论 化合物1为1对对映异构体,其中化合物1a为新化合物,命名为长春花碱A;化合物1b、2~5均为首次从该长春花属中分离得到。生物碱1b对T24细胞表现出一定抑制活性。

[Abstract]
Objective To investigate the chemical constituents of the whole herb of Catharanthus roseus (L.) G. Don and evaluate their antitumor activities. Methods The compounds were isolated and purified by silica gel column chromatography, ODS column chromatography, and HPLC, and their structures were elucidated by NMR and HR-ESI-MS spectroscopic techniques. The antitumor activities were assessed in vitro using the CCK-8 assay. Results Six alkaloids were obtained from the 95% ethanol extract of C. roseus and identified as catharosine A (1a), 2,3,3a,5-tetrahydro-1H-benzo[d]pyrrolo[2,1-b][1,3]oxazin-1-one (1b), β-carboline-1-carboxylic acid (2), 1-acetyl-β-carboline (3), β-carboline alkaloid (4), 2-(2-methyl-3-en-2-yl)-1H-indole-3-carbaldehyde (5). Conclusion Compound 1 was determined to be a pair of enantiomers, of which 1a was a new compound and named Catharosine A, while compounds 1b, 2-5 were isolated from the genus Catharanthus for the first time. Moreover, compound 1b exhibited certain inhibitory activity against T24 cells.
