目的 研究苦蘵Physalis angulata全株的化学成分及其抗炎活性。方法 综合运用多种色谱技术进行化合物的分离纯化,结合核磁共振、质谱及紫外光谱等波谱学方法鉴定其结构;采用SRB法评价化合物对小鼠巨噬细胞RAW264.7的细胞毒性,同时通过Griess法测定其体外抗炎活性。结果 从苦蘵全株的甲醇提取物中分离得到5个化合物,分别鉴定为6-5β,6β:14α,17:14β,26-三环氧-13α,15β-二羟基-1-酮-11,15-环-13,14-开环麦角甾烷-2-烯-18,26-内酯(1)、酸浆苦素B(2)、酸浆苦素F(3)、酸浆苦素XI(4)、烟曲霉三唑酯A(5)。化合物2~5具有抑制RAW264.7细胞NO生成的活性,半数抑制浓度(median inhibition concentration,IC50)值为(1.97±0.12)~(6.73±0.71)mmol/L。结论 化合物1为新化合物,命名为5,6-环氧酸浆苦素IX;化合物4、5具有显著的抗炎活性。

[Abstract]
Objective To investigate the chemical constituents from the whole plant of Physalis angulata and their anti-inflammatory activities. Methods Various chromatographic techniques were employed for the isolation and purification of compounds, and their structures were elucidated by spectroscopic methods including NMR, MS, and UV. The cytotoxicity against mouse macrophage RAW264.7 cells was evaluated using the SRB assay, while the in vitro anti-inflammatory activity was determined via the Griess assay. Results Five compounds were isolated from the methanol extract of the whole plant of P. angulata and identified as 6- 5β,6β:14α,17:14β,26-triepoxy-13α,15β-dihydroxy-1-keto-11,15-cyclo-13,14-secoergostan-2-en-18,26-olide (1), physalin B (2), physalin F (3), physalin XI (4), and aspergillus triazolate A (5). Compounds 2—5 exhibited inhibitory effects on NO production in RAW264.7 cells, with IC50 values ranging from (1.97 ± 0.12) to (6.73 ± 0.71) μmol/L. Conclusion Compound 1 is a new compound, named 5,6-epoxyphysalin IX. Compounds 4 and 5 showed significant anti-inflammatory activities.
