[摘要]
目的 研究酸模叶蓼Polygonum lapathifolium全草的化学成分。方法 利用硅胶、Sephadex LH-20和半制备高效液相色谱等分离技术进行分离纯化,通过核磁共振波谱、质谱、红外光谱等方法结合文献对分离得到的化合物进行结构鉴定。采用DPPH法、ABTS法以及羟基自由基清除方法,评价化合物的抗氧化活性。结果 从酸模叶蓼醋酸乙酯部位分离得到20个化合物,分别鉴定为2-甲基-5-n-十一烷基间苯二酚(1)、5-n-dodecylresorcinol(2)、(3S,5R)-dihydroxy-6R,7-megstigmadien-9-one(3)、植物醇(4)、山柰酚(5)、槲皮素(6)、2',4'-二羟基-6'-甲氧基查耳酮(7)、2',6'-二羟基-4'-甲氧基查耳酮(8)、花旗松素(9)、5,7,3',4'-四羟基-3-甲氧基黄酮(10)、β-谷甾醇(11)、豆甾-4-烯-6β-羟基-3-酮(12)、鞣花酸-3,3'-二甲醚(13)、没食子酸乙酯(14)、methyl 2,4,6-trihydroxybenzoate(15)、棕榈酸乙酯(16)、9(Z),12(Z)-十八烷二烯酸(17)、硬脂酸甲酯(18)、棕榈酸(19)和油酸(20)。化合物1~6、9~14、18对2,2-联苯基-1-苦基肼基(2,2-diphenyl-1-picrylhydrazyl,DPPH)和2,2-联氮二(3-乙基-苯并噻唑-6-磺酸)二铵盐 [2,2'-azinobis-(3-ethylbenzthia- zoline-6-sulphonate),ABTS] 自由基或羟基自由基具有较强的清除活性,化合物1清除DPPH自由基的IC50值为0.023 mg/mL。结论 化合物1为新的间苯二酚类化合物,命名为酸模叶蓼酚A。化合物1~3、13、15为首次从蓼属植物中分离得到。化合物1~6、9~14、18具有良好的抗氧化活性。

[Abstract]
Objective To study the chemical constituents of Polygonum lapathifolium. Methods The compounds were separated and purified by silica gel, Sephadex LH-20 column chromatography and semi-preparative HPLC. The structures were identified by NMR, MS, IR data analyses and literature data comparison. Their antioxidant activity was evaluated by using the DPPH method, the ABTS method and the hydroxyl radical scavenging method. Results A total of 20 compounds were isolated from the ethyl acetate of P. lapathifolium and elucidated as 2-methyl-5-n-undecylresorcinol (1), 5-n-dodecylresorcinol (2), (3S,5R)-dihydroxy-6R,7-megstigmadien-9-one (3), trans-phytol (4), kaempferol (5), quercetin (6), 2',4'-dihydroxy-6'-methoxychalcone (7), 2',6'-dihydroxy-4'-methoxychalcone (8), dihydroquercetin (9), 5,7,3',4'-tetrahydroxy-3-methoxyflavone (10), β-sitosterol (11), stigmaster-4-en-6β-hydroxy-3-one (12), ellagic acid-3,3'-dimethyl ether (13), ethyl gallate (14), methyl 2,4,6-trihydroxybenzoate (15), ethyl palmitate (16), 9(Z),12(Z)-octadecanedioenoic acid (17), methyl stearate (18), palmitic acid (19), oleic acid (20). Compounds 1―6, 9―14, 18 had the stronger DPPH, ABTS or OH radicals scavenging abilities and compound 1 showed significant abilities with IC50value of 0.023 mg/mL. Conclusion Compound 1 is a new resorcinol derivative, and named polygonumol A. Compounds 1―3, 13 and 15 are isolated from this genus for the first time. Compounds 1―6, 9―14, 18 exhibit significant antioxidant capacity.
