[摘要]
目的 研究安息香Benzoinum化学成分以及体外抑制肿瘤增殖活性。方法 采用MCI gel CHP-20P、正相硅胶、ODS和Sephadex LH-20柱色谱以及半制备型高效液相等色谱方法进行分离和纯化,通过红外、核磁共振、质谱等技术鉴定化合物的结构。CCK-8法评价分离得到的化合物体外抑制肿瘤增殖的活性。结果 从安息香醋酸乙酯萃取部位中分离得到3个化合物,分别鉴定为1,3-二苯甲酸酯-2-(羟基(4-羟基-3-甲氧基苯基)甲基)丙烷(1)、5-乙酰基-7-甲氧基-2-(3,4-亚甲二氧基苯基)苯骈呋喃(2)和5-甲酰基-7-甲氧基-2-(3,4-亚甲二氧基苯基)苯骈呋喃(3)。化合物2和3对人肝癌HepG2细胞增殖有抑制作用,半数抑制浓度(half maximal inhibitory concentration,IC50)值分别为41.2、52.6μmol/L。结论 化合物1和2为新化合物,命名为白花树醇酯(1)和白花树醇酮(2);化合物3为新的天然产物;其中化合物2和3对人肝癌HepG2细胞增殖表现出一定的细胞毒活性。

[Abstract]
Objective To study the chemical constituents from Benzoinum and their antitumor activities in vitro. Methods The compounds were isolated and purified by MCI gel CHP-20P, silica gel, ODS, Sephadex LH-20 column chromatography and semi-preparative HPLC. Their structures were elucidated by spectroscopic analysis including IR, NMR and MS. CCK-8 assay was used to evaluate the inhibitory activity of the isolated compounds on tumor proliferation in vitro. Results A total of three compounds, 1,3-dibenzoate-2-(hydroxy(4-hydroxy-3-methoxyphenyl) methyl)propane (1), 5-acetyl-7-methoxy-2-(3,4-methylenedioxyphenyl) benzofuran (2), and 5-formyl-7-methoxy-2-(3,4-methylenedioxyphenyl)benzofuran (3), were isolated from the ethyl acetate extraction site of Benzoinum. Compounds 2 and 3 exhibited the inhibitory effects on HepG-2 cells with half maximal inhibitory concentration (IC50) values of 41.2, 52.6 μmol·L−1, respectively. Conclusion Compounds 1 and 2 are new compounds named styratonate (1) and styratonone (2), and compound 3 is a new natural product, among which compounds 2 and 3 exhibit certain cytotoxic activities on the proliferation of HepG2 cells.
