[摘要]
目的 研究消癥丸对四氯化碳诱导的小鼠肝纤维化的治疗作用。方法C57BL/6小鼠随机分为对照组、模型组和消癥丸(0.91 g/kg)组,每组12只。采用连续6周ip 15%四氯化碳橄榄油溶液(2 mL/kg)制备小鼠肝纤维化模型,于造模第3周给予药物干预。给药结束后,检测血清中丙氨酸氨基转移酶(alanine aminotransferase,ALT)和天冬氨酸氨基转移酶(aspartate aminotransferase,AST)活性及肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-6(interleukin-6,IL-6)水平;测定肝组织超氧化物歧化酶(superoxide dismutase,SOD)活性及丙二醛(malondialdehyde,MDA)、羟脯氨酸(hydroxyproline,Hyp)含量;采用苏木素-伊红(HE)染色观察肝组织病理变化,天狼猩红染色观察肝组织胶原沉积情况;采用Western blotting检测肝组织Ι型胶原蛋白(type I collagen,Col-Ⅰ)、α-平滑肌肌动蛋白(α-smooth muscle actin,α-SMA)、TNF-α、Smad、转化生长因子-β(transforming growth factor-β,TGF-β)表达。结果 与对照组比较,模型组小鼠血清中ALT、AST活性及TNF-α、IL-6水平明显升高(P<0.05);肝组织SOD活性明显降低(P<0.05),MDA和Hyp含量明显升高(P<0.05);肝细胞排列紊乱,可见炎性细胞浸润,肝组织胶原沉积明显;肝组织Col-Ⅰ、α-SMA、TNF-α、Smad、TGF-β蛋白表达水平显著升高(P<0.05)。经消癥丸干预后,以上指标均得到显著改善(P<0.05)。结论 消癥丸可有效改善四氯化碳诱导的小鼠肝纤维化。

Objective To study the therapeutic effect of Xiaozheng Pills (消癥丸) on carbon tetrachloride-induced liver fibrosis in mice. Methods C57BL/6 mice were randomly divided into control group, model group, and Xiaozheng Pills (0.91 g/kg) group, with 12 mice in each group. A mouse liver fibrosis model was prepared using 15% carbon tetrachloride olive oil solution (2 mL/kg) for six consecutive weeks, and drug intervention was administered in 3rd week of modelling. After administration, activities of alanine aminotransferase (ALT), aspartate aminotransferase (AST) and levels of tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6) in serum were measured; The activity of superoxide dismutase (SOD) and contents of malondialdehyde (MDA), hydroxyproline (Hyp) in liver tissue were measured; Hematoxylin-eosin (HE) staining was used to observe pathological changes in liver tissue, and sirius red staining was used to observe collagen deposition in liver tissue; Western blotting was used to detect expressions of type I collagen (Col-I), α-smooth muscle actin (α-SMA), TNF-α, Smad and transforming growth factor-β (TGF-β) in liver tissue. Results Compared with control group, the activities of ALT, AST and levels of TNF-α, IL-6 in serum of mice in model group were significantly increased (P < 0.05), SOD activity in liver tissue was significantly reduced (P < 0.05), while the contents of MDA and Hyp were significantly increased (P < 0.05); The arrangement of liver cells was disordered, with visible infiltration of inflammatory cells, deposition of collagen in liver tissue was significant; The expression levels of Col-Ⅰ, α-SMA, TNF-α, Smad and TGF-β proteins in liver tissue were significantly increased (P < 0.05). After intervention with Xiaozheng Pills, the above indicators were significantly improved (P < 0.05). Conclusion Xiaozheng Pills can effectively improve liver fibrosis induced by carbon tetrachloride in mice.
