目的 研究杭白芷Angelica dahurica var. formosana的化学成分。方法 利用各种色谱分离技术对临床常用中药白芷中的香豆素类成分进行研究,结合核磁共振波谱对分离得到的单体化合物进行结构鉴定,并对所有的化合物进行了抗炎活性筛选。结果 从白芷乙醇提取物中共得到22个香豆素类化合物,分别鉴定为 (4S)-4,8-dihydroxy-5-prenyl-3,4-dihydro-furocoumarin(1)、花椒毒酚(2)、花椒毒素(3)、佛手柑内酯(4)、异茴芹灵(5)、异去甲基呋喃羽叶芸香素(6)、异欧前胡素(7)、欧前胡素(8)、珊瑚菜素(9)、比克白芷素(10)、水合氧化前胡素(11)、别欧前胡素(12)、异氧化前胡内酯(13)、新白当归脑(14)、dahurin B(15)、仲-O-β-D-吡喃葡萄糖基白当归素(16)、叔-O-β-D-吡喃葡萄糖基- (R)-白当归素(17)、7-去甲软木花椒素(18)、紫花前胡苷元(19)、紫花前胡苷(20)、印度榅桲苷(21)、marmesin 4'-O-β-D-apiofuranosyl- (1-6)-O-β-D-glucopyranoside(22)。结论 化合物1为新化合物,命名为 (4S)-3,4-二氢白芷素B;化合物2、11对脂多糖(lipopolysaccharide,LPS)诱导的小鼠巨噬细胞RAW 264.7的一氧化氮(NO)产生有一定抑制活性,半数抑制浓度(median inhibition concentration,IC50)分别为(32.8±0.8)µmol/L和(94.5±1.9)µmol/L。

[Abstract]
Objective To investigate the chemical constituents of Angelica dahurica var. formosana. Methods The chemical constituents of of Angelica Dahurica Radix were studied by using various chromatographic techniques, and their structures were established on the basis of NMR spectra, and all compounds were screened for anti-inflammatory activity. Results A total of 22 coumarins were isolated and identified from the ethanol extract of A. dahurica var. formosana, including (4S)-4,8-dihydroxy-5-prenyl-3,4-dihydro-furocoumarin (1), xanthotoxol (2), xanthotoxin (3), bergapten (4), isoimpinellin (5), isodemethylfuropinarine (6), isoimperatorin (7), imperatorin (8), phellopterin (9), byakangelicin (10), oxypeucedanin hydrate (11), alloimperatorin (12), isooxypeucedanin (13), neobyakangelicol (14), dahurin B (15), sec-O-β-D-glucopyranosyl-byakangelicin (16), tert-O-β-D-glucopyranosyl-(R)-byakangelicin (17), demethylsuberosin (18), decursin (19), nodakenin (20), marmesinin (21), marmesin 4'-O-β-D-apiofuranosyl-(1-6)-O-β-D-glucopyranoside (22). Conclusion Compound 1 was identified as a new compound, named as (4S)-angelicadin B. Compounds 2 and 11 showed inhibitory activity against the nitric oxide production in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophage cells with median inhibition concentration (IC50) values of (32.8 ±0.8) µmol/L and (94.5 ±1.9) µmol/L, respectively.
