目的 基于UPLC-Q-Exactive Orbitrap-MS/MS联用技术研究中药复方五神汤中的化学成分,归纳总结不同类型化合物的裂解规律。方法 采用ShiM-pack XR-ODS C18色谱柱(75 mm×3.0 mm,2.2 μm),流动相为乙腈(B)-水(A),梯度洗脱;体积流量0.5 mL/min;柱温30 ℃;电喷雾离子源(ESI),在正、负离子模式下对五神汤提取液进行质谱检测。结果 从五神汤提取液中分析鉴定出95个化合物,包括27个黄酮类、15个酚酸类、11个环烯醚萜类、5个香豆素类、5个生物碱类、4个苯乙醇苷类以及28个其他类化合物,并对金丝桃苷、木犀草苷、新绿原酸、京尼平苷酸、马钱苷酸、毛蕊花糖苷、异麦角甾苷、秦皮甲素等主要化学成分进行裂解规律解析。结论 建立的方法准确可靠,揭示了中药复方五神汤的潜在有效成分。

[Abstract]
Objective To study the compounds in Wushen Decoction (五神汤) based on UPLC-Q-Exactive Orbitrap-MS/MS technology, and to summarize the cracking laws of different types of compounds. Methods The chromatography was performed on ShiM-pack XR-ODS C18 column (75 mm×3.0 mm, 2.2 μm) with acetonitrile (B)-water (A) as mobile phase and gradient elution. Volume flow rate was 0.5mL/min and column temperature was 30 ℃. An electrospray ion source (ESI) was used to detect the extract of Wushen Decoction by mass spectrometry in positive and negative ion modes. Results A total of 95 compounds, including 27 flavonoids, 15 phenolic acids, 11 iridoid glycosides, 5 coumarins, 5 alkaloids, 4 phenylethanol glycosides and other 28 compounds, were identified from the extract of Wushen Decoction. The cracking laws of their main chemical components such as hyperoside, luteolin, nechlorogenic acid, geniposide, loganin, verbascoside, isoacteoside and esculin were analyzed. Conclusion The established method is accurate and reliable, which reveals the potential effective components of the traditional Chinese medicine compound Wushen Decoction.
