[摘要]
目的 对缬草Valeriana officinalis水提取物中的化学成分进行研究。方法 运用正相硅胶柱色谱、ODS反相柱色谱、制备高效液相色谱等分离手段进行分离纯化,采用核磁共振、质谱等多种波谱学技术鉴定化合物的化学结构,并采用Griess法评价其对脂多糖诱导的小胶质细胞BV-2神经炎症的抑制活性。结果 从缬草水提取物中分离得到了12个化合物,分别为tinnevellin-8-O-β-D-glucopyranoside(1)、大黄素-8-O-β-D-吡喃葡萄糖苷(2)、(2R,3S)-肥牛木素(3)、左旋橄榄素-9'-O-β-D-吡喃葡萄糖苷(4)、左旋马尾松树脂醇-3a-O-β-D-吡喃葡萄糖苷(5)、金色酰胺醇乙酸酯(6)、松脂素(7)、8-羟基松脂素(8)、松脂素-4-O-β-D-吡喃葡萄糖苷(9)、8,8'-二羟基松脂素-4-O-β-D-吡喃葡萄糖苷(10)、(2S)-橙皮苷(11)和香蜂草苷(12)。结论化合物1~4和6均为首次从缬草属植物中分离得到。其中,化合物3~4、6、9、11~12在不影响BV-2的细胞活力前提下,可以抑制脂多糖诱导的BV-2细胞神经炎性因子一氧化氮(NO)的产生。

[Abstract]
Objective To study the chemical constituents of the water-soluble extract of Valeriana officinalis.Methods Silica gel and ODS column chromatography, preparative high-performance liquid chromatography were applied for isolation of constituents from the water-soluble extract. NMR, MS and other related spectroscopic analyses were used to identify their structures, and a Griess method was used to evaluate their inhibitory activity on lipopolysaccharide-induced inflammation of BV-2 cells.Results Twelve compounds were isolated and identified as tinnevellin-8-O-β-D-glucopyranoside (1), emodin-8-O-β-D-glucopyranoside (2), (2R,3S)-ceplignan (3), (-)-olivil-9'-O-β-D-glucopyranoside (4), (-)-massoniresinol-3a-O-β-D-glucopyranoside (5), aurantiamide acetate (6), pinoresinol (7), 8-hydroxypinoresinol (8), pinoresinol-4-O-β-D-glucopyranoside (9), prinsepiol-4-O-β-Dglucopyranoside (10), (2S)-hesperidin (11) and didymin (12).Conclusion Compounds 1-4 and 6 are firstly isolated from the genus Valeriana. Compound 3-4、 6、 9、 11-12 showed inhibition on the production of neuroinflammatory factor NO induced by LPS in BV-2 cells, without any detectable influence on the cell viabilities.
