[摘要]
目的 研究毛华菊Chrysanthemum vestitum的化学成分及体外肿瘤细胞毒活性。方法 采用溶剂提取法、Diaion HP-20柱色谱、Sephadex LH-20葡聚糖凝胶柱色谱、硅胶柱色谱及制备高效液相色谱等色谱方法,对毛华菊氯仿部位的化学成分进行分离纯化,并利用现代波谱技术对已分离得到的化合物进行结构鉴定。MTT法测定各化合物体外对人肝癌HepG2细胞增殖的抑制活性。结果 从毛华菊的氯仿部位分离得到3个化合物,分别鉴定为(1S,5R,6R,7R,8S,10S)-8-乙酰氧基-愈创木-3,11(13)-二烯-2-酮-12,6-内酯(1)、(1S,5R,6R,7R,8S,10S,11S)-8-乙酰氧基-愈创木-3-烯-2-酮-12,6-内酯(2)和2α-(2',4'-hexadiynoyl)-1,6-dioxaspiro[4,5]-deca-3-ene(3)。结论 化合物2为1个新的愈创木内酯,命名为毛华菊内酯A,其对HepG2细胞增殖没有明显的抑制作用;化合物1的氢谱全谱、碳谱以及绝对构型为首次报道,它对HepG2细胞具有较强的体外抑制作用,半数抑制浓度(median inhibition concentration,IC50)值为(5.95±0.19)μmol/L;化合物3为首次从毛华菊中分离得到。

[Abstract]
Objective To investigate the chemical constituents from Chrysanthemum vestitum and their anti-tumor activities. Methods The chemical constituents from C. vestitum were isolated and purified by solvent extraction, Diaion HP-20, Sephadex LH-20, Silica gel and preparative high performance liquid chromatography. Their structures were elucidated based on their spectroscopic data. The antitumor activities of the isolated compounds were evaluated by using MTT assay. Results Three compounds were isolated from chloroform extract of C. vestitum and identified as (1S,5R,6R,7R,8S,10S)-2-oxo-8- acetoxy-guaia-3,11(13)-dien-12,6-olide (1), (1S,5R,6R,7R,8S,10S,11S)-2-oxo-8-acetoxy-guaia-3-en-12,6-olide (2) and 2α-(2',4'- hexadiynoyl)-1,6-dioxaspiro[4,5]-deca-3-ene (3). Conclusion Compound 2 is a new guaianolide named as vestitlactone A which had no significant inhibitory effect on the HepG2 cell proliferation; compound 1 exhibited inhibitory activity against HepG2 cell line with IC50 value of (5.95 ±0.19) μmol/Land compound 3 was isolated from this plant for the first time.
