目的 建立基于超高效液相色谱-三重四极杆串联质谱(UPLC-QQQ-MS/MS)技术的工业大麻中6种大麻素类成分的定量分析方法,并利用该方法对9个火麻仁基原植物工业大麻植株3个不同部位(雌花、苞片、叶)中的大麻素类成分含量进行研究,为合理合法开发利用工业大麻,甄选商业化品种以及选育符合我国法规的高大麻二酚(cannabidiol,CBD)含量、低四氢大麻酚(tetrahydrocannabinol,THC)含量品种奠定基础。方法 采用液质联用技术:Agilent Eclipse Plus C18色谱柱(50 mm×2.1 mm,1.8 μm),流动相为0.1%甲酸水溶液(A)-甲醇(B),梯度洗脱,体积流量0.3 mL/min,多反应监测模式。结果 以工业大麻中6种大麻素类成分大麻二酚、四氢大麻酚、大麻萜酚(cannabigerol,CBG)、大麻二醇酸(cannabidiolic acid,CBDA)、大麻萜酚酸(cannabigerolic acid,CBGA)和四氢大麻酚酸(tetrahydrocannabinolic acid,THCA)为研究对象,建立了基于UPLC-QQQ-MS/MS的定量方法。方法学验证结果显示:6种大麻素类成分线性良好,R2为0.999 1~1.000 0,准确度为0.29%~2.80%,精密度为0.84%~2.74%,加样回收率为99.36%~102.88%。样品分析结果显示:花和苞片中的6种大麻素类成分含量显著高于叶片中的含量,不同品种大麻中6种大麻素类成分在花、苞片和叶中的分布具有特异性。结论 基于UPLC-QQQ-MS/MS所建立的大麻中6种大麻素类成分的定量分析方法,灵敏度高,准确性好,方法简单,适用于工业大麻中6种大麻素类成分的含量测定。

[Abstract]
Objective To establish a quantitative analysis method for six cannabinoids in hemp based on ultra-performance liquid chromatography coupled to triple-quadrupole tandem mass spectrometer (UPLC-QQQ-MS/MS) technology, and use this method to study the content of cannabinoids in three different parts (female flowers, bracts, and leaves) of cannabis plants in nine international mainstream hemp. It lays a foundation for the rational and legal development and utilization of medicinal cannabis, the selection of commercial varieties and the breeding of varieties with high cannabidiol (CBD) content and low tetrahydrocannabinol (THC) content in accordance with Chinese laws and regulations. Methods The chromatographic separation was performed on an Agilent Eclipse Plus C18 column (50 mm x2.1 mm, 1.8 μm) with a gradient elution, the mobile phase was 0.1% formic acid aqueous solution (A)-methanol (B), and the flow rate was set at 0.3 mL/min; Multiple reaction monitoring (MRM) mode was used for the quantitative analysis of six cannabinoids in hemps. Results Taking six CBD, THC, cannabigerol (CBG), cannabidiolic acid (CBDA), cannabigerolic acid (CBGA), and tetrahydrocannabinolic acid (THCA) in hemp as the research object, a quantitative method based on UPLC-QQQ-MS/MS was developed. The method verification results showed that the six cannabinoids had good linearity, with R2 ranging from 0.9991 to 1.000, accuracy ranging from 0.29% to 2.80%, precision ranging from 0.84% to 2.74%, and the sample recovery rate was 99.36% to 102.88%. The sample analysis results showed that the content of six cannabinoids in flowers and bracts was significantly higher than that in leaves, and the distribution of six cannabinoids in different varieties of cannabis in flowers, bracts and leaves was specific. Conclusion The quantitative analysis method established by UPLC-QQQ-MS/MS has high sensitivity, good accuracy and simple method, which is suitable for the determination of cannabinoids in hemp.
