目的 采用超高效液相色谱-四极杆-飞行时间质谱联用技术(UPLC-Q/TOF-MS)对痹祺胶囊化学物质组和入血成分进行辨识研究,初步阐明其可能的药效物质基础。方法 采用Acquity BEH C18色谱柱(100 mm×2.1 mm,1.7 μm),流动相为乙腈(A)-0.1%甲酸水溶液(B),梯度洗脱,体积流量为0.2 mL/min,柱温为30℃,进样量为5 μL。采用电喷雾离子源,正、负离子模式扫描。离子源温度为110℃,锥孔电压为30 V,锥孔气体积流量为50 L/h;脱溶剂用氮气,脱气温度为350℃,体积流量为800 L/h;扫描范围为m/z 50~2000。结果 在痹祺胶囊样品溶液中鉴定出280个成分,在给药大鼠血浆样品中鉴定出81个化学成分,包括59个原型成分和22个代谢产物。结论 对痹祺胶囊及其入血成分进行了较全面地研究,初步阐明痹祺胶囊的药效物质基础,为痹祺胶囊的后续研究提供了理论基础。。

[Abstract]
Objective To identify the chemical substance and blood components of Biqi Capsules (痹祺胶囊), and preliminarily clarify its possible medicinal substances basis by ultra-high performance liquid chromatography-quadrupole-time-of-flight mass spectrometry (UPLC-Q/TOF-MS). Methods The separation was performed on Acquity BEH C18 chromatographic column (100 mm×2.1mm, 1.7 μm) with mobile phase of acetonitrile (A)-0.1% formic acid aqueous solution (B) for gradient elution at volume flow rate of 0.2 mL/min under column temperature of 30℃. The injection volume was 5 μL. The electrospray ion source was used for scanning in positive and negative ion modes. Ion source temperature was 110℃, cone voltage was 30 V, cone gas volume flow was 50 L/h, nitrogen degassing temperature was 350℃, degassing volume flow was 800 L/h, scanning range m/z 50-2000. Results A total of 280 components were identified in Biqi Capsule sample solution, and 81 chemical components were identified in plasma samples, including 59 prototype components and 22 metabolites. Conclusion This experiment conducted a more comprehensive study of Biqi Capsules and their blood components, and initially clarified the material basis of the efficacy of Biqi Capsules, and provided a theoretical basis for the follow-up research of Biqi Capsules.
