目的 建立吴茱萸药材HPLC定量指纹图谱,为其基原鉴别及质量评价提供依据。方法 针对29批3种基原吴茱萸药材,建立吴茱萸HPLC定量指纹图谱,并通过HPLC-ESI-MS技术对指纹图谱中的色谱峰进行鉴定,采用《中药色谱指纹图谱相似度评价系统》计算相似度,同时运用化学模式识别技术对3种基原吴茱萸药材进行区分,并对吴茱萸中指标性成分进行含量测定。结果 建立了吴茱萸药材的HPLC定量指纹图谱,标定13个共有峰,并通过液质联用进行了共有峰的鉴定。聚类分析和正交最小偏二乘判别分析实现了3种吴茱萸基原药材的鉴别。3种基原吴茱萸药材中吴茱萸碱、吴茱萸次碱、去氢吴茱萸碱和水仙苷4个指标性成分含量存在一定差异。结论 通过吴茱萸HPLC定量指纹图谱与化学模式识别技术相结合,可以实现对不同基原的吴茱萸药材的有效区分,为吴茱萸药材的质量控制提供参考依据。

[Abstract]
Objective To establish HPLC quantitative fingerprint of Wuzhuyu (Evodia rutaecarpa), so as to provide basis for its species differentiation and quality evaluation. Methods The HPLC quantitative fingerprint of 29 batches of E.rutaecarpa was established from three species of E. rutaecarpa. The chromatographic peaks in the fingerprint were identified by HPLC-ESI-MS technology. The similarity was calculated by the similarity evaluation system of traditional Chinese medicine chromatographic fingerprint. Chemical pattern recognition technology was used to distinguish three kinds of E. rutaecarpa, and the content of the index components in E. rutaecarpa was determined. Results In this study, the HPLC quantitative fingerprint of E. rutaecarpa was established, 13 common peaks were identified, and the structure of common peaks was identified by liquid chromatography-mass spectrometry. Cluster analysis and orthogonal partial least squares discriminant analysis were used to identify the three species of E. rutaecarpa. The contents of evodiamine, rutaecarpine, dehydroaevodiamine and narcissoside in E. rutaecarpa from three species are different. Conclusion The effective distinguishment of different species of E. rutaecarpa can be achieved through the combination of HPLC quantitative fingerprint and chemical pattern recognition technology, which will provide reference for the quality control of E. rutaecarpa.
