[摘要]
目的 研究参芪扶正注射液对微小RNA-29b(microRNA-29b,miR-29b)和B淋巴细胞瘤2(B-cell lymphoma 2,Bcl-2)表达的影响,探究参芪扶正注射液逆转胰腺癌多药耐药的可能机制。方法 将miR-29b模拟物(miR-29b mimics)和miR-29b抑制剂(miR-29b inhibitors)转染至人胰腺癌5-氟尿嘧啶(5-fluorouracil,5-Fu)耐药细胞株Patu8988/5-Fu,采用qRT-PCR检测参芪扶正注射液(5、10、20 μL/mL)对Patu8988/5-Fu细胞miR-29b mRNA表达的影响;采用MTT法检测miR-29b mimics和miR-29b inhibitors对Patu8988细胞活力的影响;采用Western blotting法检测miR-29b mimics和miR-29b inhibitors对Patu8988/5-Fu细胞Bcl-2蛋白表达的影响。结果 与对照组比较,参芪扶正注射液组Patu8988/5-Fu细胞中miR-29b mRNA表达水平显著升高(P<0.05、0.01、0.001);miR-29b mimics组Patu8988/5-Fu细胞miR-29b mRNA表达水平显著升高(P<0.001),miR-29b inhibitors组miR-29b mRNA表达水平显著降低(P<0.01);miR-29b mimics组Patu8988细胞活力显著降低(P<0.05),miR-29b inhibitors组Patu8988细胞活力显著升高(P<0.01);miR-29b mimics组Patu8988/5-Fu细胞Bcl-2蛋白表达水平显著降低(P<0.05),miR-29b mimics+参芪扶正注射液组Bcl-2蛋白表达水平进一步降低(P<0.05);miR-29b inhibitors组Patu8988/5-Fu细胞Bcl-2蛋白表达水平显著升高(P<0.05),miR-29b mimics+参芪扶正注射液组Bcl-2蛋白水平无明显变化。结论 miR-29b通过下调Bcl-2蛋白表达抑制Patu8988细胞增殖,参芪扶正注射液可能通过调控miR-29b/Bcl-2通路从而逆转肿瘤多药耐药。

[Abstract]
Objective To study the effect of Shenqi Fuzheng Injection (参芪扶正注射液) on expression of microRNA-29b (miR-29b) and B-cell lymphoma 2 (Bcl-2), and explore the possible mechanisms of Shenqi Fuzheng Injection on reversing multidrug resistance in pancreatic cancer. Methods miR-29b mimics and miR-29b inhibitors were transfected into human pancreatic cancer 5-fluorouracil (5-Fu) resistant cell line Patu8988/5-Fu, qRT-PCR was used to detect the effect of Shenqi Fuzheng Injection (5, 10, 20 μL/mL) on miR-29b mRNA expression in Patu8988/5-Fu cells; MTT method was used to detect the effect of miR-29b mimics and miR-29b inhibitors on viability of Patu8988 cells; Western blotting method was used to detect the effect of miR-29b mimics and miR-29b inhibitors on Bcl-2 protein expression in Patu8988/5-Fu cells. Results Compared with control group, miR-29b mRNA level in Patu8988/5-Fu cells in Shenqi Fuzheng Injection group was significantly increased (P < 0.05, 0.01, 0.001); miR-29b mRNA level of Patu8988/5-Fu cells in miR-29b mimics group was significantly increased (P < 0.001), miR-29b mRNA level in miR-29b inhibitors group was significantly decreased (P < 0.01); Viability of Patu8988 cells in miR-29b mimics group was significantly decreased (P < 0.05), viability of Patu8988 cells in miR-29b inhibitors group was significantly increased (P < 0.01); Bcl-2 protein expression of Patu8988/5-Fu cells in miR-29b mimics group was significantly reduced (P < 0.05), Bcl-2 protein expression in miR-29b mimics + Shenqi Fuzheng Injection was further reduced (P < 0.05); Bcl-2 protein level of Patu8988/5-Fu cells in miR-29b inhibitors group was significantly increased (P < 0.05), Bcl-2 protein expression in miR-29b mimics + Shenqi Fuzheng Injection group had no significant change. Conclusion miR-29b inhibits the proliferation of Patu8988 cells by down-regulating Bcl-2 expression, Shenqi Fuzheng Injection may reverse tumor multidrug resistance by regulating miR-29b/Bcl-2 pathway.
