[摘要]
目的 研究苗药血人参(茸毛木蓝Indigofera stachyodes干燥根)中的化学成分。方法 采用硅胶、ODS、Sephadex LH-20柱色谱和半制备高效液相色谱技术对血人参中的化学成分进行分离纯化,通过其理化性质和NMR、MS等谱学数据进行结构鉴定。结果 从血人参乙醇提取物的醋酸乙酯萃取部位分离鉴定了24个黄酮类化合物,分别鉴定为芒柄花素(1)、7-羟基-3',4'-二甲氧基异黄酮(2)、染料木素(3)、毛蕊异黄酮(4)、大豆素(5)、farnisin(6)、3',4',7-三羟基黄酮(7)、木犀草素(8)、漆黄素(9)、甘草素(10)、erycibenin D(11)、3,4',7-三羟基二氢黄酮(12)、二氢山柰酚(13)、黄颜木素(14)、6,4'-二羟基-3'-甲氧基橙酮(15)、硫黄菊素(16)、(–)-3,4',7-三羟基-3'-甲氧基黄烷(17)、儿茶素(18)、表儿茶素(19)、(‒)-epicatechin-3-O-p-hydroxybenzoate(20)、美迪紫檀素-3-O-β-D-葡萄糖苷(21)、2',4'-二甲氧基-3'-羟基异黄烷-6-O-β-D-葡萄糖苷(22)、7-羟基色原酮(23)和5,7-二羟基色原酮(24)。结论 化合物2、5~7、11~15、17、20~24为首次从木蓝属植物中分离得到,化合物1、10和16为首次从血人参中分离得到。化合物3、7、8、10、15和16能够抑制LPS诱导巨噬细胞RAW 264.7释放NO,其半数抑制浓度(IC50)在7.3~27.3μmol/L。

[Abstract]
Objective To investigate the chemical constituents from the roots of Miao medicine Indigofera stachyodes. Methods The compounds were isolated and purified by various column chromatographies and their structures were identified by physiochemical properties and spectroscopic data. Results Twenty-four compounds were isolated from the EtOAc extract from the roots of I. stachyodes and identified as formononetin (1), 7-hydroxy-3ʹ,4ʹ-dimethoxyisoflavone (2), genistein (3), calycosin (4), daidzein (5), farnisin (6), 3ʹ,4ʹ,7-trihydroxyflavone (7), luteolin (8), fisetin (9), liquiritigenin (10), erycibenin D (11), 3,4ʹ,7-trihydroxyflavanone (12), dihydrokaempferol (13), fustin (14), 6,4ʹ-dihydroxy-3ʹ-methoxyaurone (15), sulfuretin (16), (–)-3,4ʹ,7-trihydroxy-3ʹ-methoxyflavan (17), catechin (18), epicatechin (19), (‒)-epicatechin-3-O-p-hydroxybenzoate (20), medicarpin-3-O-β-D-glucoside (21), 2ʹ,4ʹ-dimethoxy-3ʹ-hydroxy-isoflavan-6-O-β-D-glucoside (22), 7-hydroxychromone (23) and 5,7-dihydroxychromone (24). Conclusion Compounds 2, 5—7, 11—15, 17, and 20—24 are firstly isolated from the genus Indigofera, compounds 1, 10 and 16 are obtained from the roots of I. stachyodes for the first time. Compounds 3, 7, 8, 10, 15 and 16 exhibited significant inhibition of nitric oxide production in lipopolysaccharide-stimulated RAW 264.7 cells with IC50 values in the range of 7.3—27.3 μmol/L.
