[摘要]
目的 采用高效液相色谱串联四极杆飞行时间质谱联用技术(HPLC-Q-TOF-MS/MS)对七味通痹口服液中的化学成分进行定性表征。方法 色谱柱为Agilent ZORBAX SB-C18(250 mm×4.6 mm,5 μm),以甲醇-0.1%甲酸水溶液为流动相,梯度洗脱,体积流量1.0 mL/min,柱温25 ℃;采用电喷雾离子源,正、负离子全扫描模式下采集数据,根据精确质荷比、二级碎片离子信息,结合对照品和文献数据鉴定七味通痹口服液中的化学成分。结果 共指认出七味通痹口服液制剂中72个化学成分,包括生物碱类、黄酮类、氨基酸类、酚酸类及苯丙素类等,其中18个化合物经对照品比对。结论 该研究为七味通痹口服液的药效物质基础及质量控制的深入研究奠定了基础。

[Abstract]
Objective To study the chemical constituents of Qiwei Tongbi Oral Liquid (七味通痹口服液) by using high performance liquid chromatography-quadrupole time-of-flight mass spectrometry (HPLC-Q-TOF-MS/MS). Methods The analysis was performed on Agilent ZORBAX SB-C18 (250 mm×4.6 mm, 5 μm) column. The mobile phase consisted of methanol and 0.1% formic acid aqueous solution was used as gradient elution. The flow rate was 1.0 mL/min and column temperature was 25 ℃. The mass spectrometry was acquired in both negative and positive ion modes by using electron spray ionization (ESI). These components were further analyzed based on high-resolution mass-to-charge ratios, fragment ion species and other information combined with reference substance and literature data. Results A total of 72 compounds were identified and predicted, including alkaloids, flavonoids, amino acid, phenolic acids and phenylpropanoids, of which 18 compounds were verified by reference substances. Conclusion This study lay the foundation for elucidating its active components, and provide reference for the quality control of Qiwei Tongbi Oral Liquid.
