目的 通过体外实验研究β-羟基异戊酰紫草素(β-HIVS)对人肺腺癌A549细胞生长的抑制作用,并对其机制进行初步探讨。方法 体外培养A549细胞,分为对照组和不同浓度的β-HIVS组。采用CCK-8法、流式细胞术、Transwell小室法检测β-HIVS对A549细胞增殖、周期、凋亡、侵袭和迁移的影响;实时荧光定量PCR技术和Western blotting法检测β-HIVS处理后A549细胞p53、Bcl-2的mRNA和蛋白表达情况。结果 β-HIVS能抑制A549细胞的增殖;诱导A549细胞凋亡;阻滞细胞由G0/G1期向S期的发展;抑制细胞侵袭和迁移;呈浓度依赖性地上调p53蛋白和mRNA的表达,下调Bcl-2蛋白和mRNA的表达。结论 β-HIVS可显著抑制A549细胞的增殖、侵袭和迁移,并诱导细胞凋亡,其机制可能与上调p53的表达和下调Bcl-2的表达有关。

[Abstract]
Objective To study the effects of β-hydroxyisovaleryl shikonin (β-HIVS) on the growth of human lung cancer A549 cells in vitro, and to explore its mechanism. Methods A549 cells were cultured in vitro and divided into control group and β-HIVS groups. The effect of β-HIV on cell viability, cycle and apoptosis, invasion and migration of cells were studied by CCK-8 assay, flow cytometric and Transwell assay, respectively. The expression of p53 and Bcl-2 protein, and mRNAs were assessed by Western blotting and quantitative real-time PCR analysis.Results β-HIV could inhibit the proliferation of A549 cells, induce the apoptosis of A549 cells, block the G0/G1 phase progression to S phase, inhibit cell invasion and migration, upregulate the expression of p53 protein and mRNA in a concentration-dependent manner, and down-regulate the expression of Bcl-2 protein mRNA expression. Conclusion β-HIV can significantly inhibit the proliferation, invasion and migration of A549 cells and induce the apoptosis by regulating the expression of p53 and Bcl-2.
