目的 研究香加皮中提取的宝藿苷-Ⅰ对人食管癌细胞Eca-109的抑制作用及其对Wnt/β-catenin信号转导通路的影响. 方法 RT-PCR技术检测不同质量浓度宝藿苷-Ⅰ作用48 h后,细胞β-catenin、Cyclin D1、Survivin m RNA表达的变化,采用流式细胞术分析Eca-109细胞β-catenin、Cyclin D1、Survivin蛋白的表达. 结果 宝藿苷-I(25、50μg/mL)作用48 h后,Eca-109细胞β-catenin、Cyclin D1、Survivin mRNA及蛋白表达水平明显下降(P<0.01),12.5μg/mL处理组细胞的β-catenin、Cyclin D1、Survivin mRNA及其蛋白表达水平与对照组相比无显著差异. 结论 宝藿苷-I可能通过下调β-catenin、Cyclin D1、Survivin mRNA及其蛋白表达,影响Wnt/β-catenin信号转导通路,发挥抑制Eca-109细胞增殖作用.

[Abstract]
Objective To study the effect of baohuoside-I extracted from Periplocae Cortex on proliferation and Wnt/β-catenin signaling pathway of human esophageal carcinoma cell Eca-109. Methods The expressions of β-catenin, Cyclin D1, and Survivin protein in Eca-109 cells were measured with flow cytometry (FCM). The expressions of β-catenin, Cyclin D1, and Survivin mRNA were detected by RT-PCR. Results After treatment with 25 and 50 μg/mL of baohuoside-I for 48 h, the expression levels of β-catenin, Cyclin D1, Survivin mRNA, and protein were decreased significantly (P<0.01), but with 12.5 μg/mL of baohuoside-I the expression level was not decreased significantly compared with the control group. Conclusion Baohuoside-I from Periplocae Cortex could inhibit the proliferation of Eca-109 cells. This effect associats with down-regulation expression of β-catenin, Cyclin D1, Survivin, and their proteins, which affects on the Wnt/β-catenin signaling pathway.
