[摘要]
目的建立款冬花药材中槲皮素和山柰素的定量测定方法,测定不同来源的药材中槲皮素和山柰素的量,评价产地、生长方式及炮制对款冬花中槲皮素和山柰素量的影响;并建立款冬花中黄酮醇苷类成分指纹图谱,为款冬花质量控制的制定提供依据。方法采用HPLC建立款冬花药材中槲皮素和山柰素的定量测定方法,色谱条件:Diamonsil C18(200 mm×4.6 mm,5μm),流动相为乙腈-水(77∶23),检测波长:365 nm。采用HPLC法建立款冬花药材中黄酮醇苷类成分指纹图谱,色谱条件:ODS-BP C18(200 mm×4.6 mm,5μm);流动相:乙腈-0.1%H3PO4水溶液梯度洗脱;检测波长为365 nm。结果不同产地,炮制品与生品之间,其槲皮素和山柰素的量差异较大,不同生长方式的药材之间无明显差异;不同产地及批次的款冬花药材的指纹图谱相似度较高。结论本研究建立的槲皮素和山柰素的定量测定方法简便、准确、重现性好,可用于款冬花药材中黄酮醇苷类成分的质量控制;不同来源的款冬花药材中的槲皮素和山柰素的量有明显差异;所建立的黄酮醇苷类成分的指纹图谱方法操作性强、重现性好,可作为款冬花黄酮醇苷类成分指纹图谱研究的基础。

Objective To establish the method for quantitative analysis of quercetin and kaempferide in Flos Farfarae,to determine the contents of quercetin and kaempferide in Flos Farfarae from various habitats,to evaluate the effects of habitats,growth way,and processing on the contents of quercetin and kaempferide in Flos Farfarae,and to establish the fingerprint of flavone glycosides in Flos Farfarae,so as to provide the information for quality control of Flos Farfarae.Methods Quantification of quercetin and kaempferide in Flos Farfarae was performed by HPLC on Diamonsil C18 column(200 mm×4.6 mm,5 μm) using acetonitrile-H2O(77∶23) as mobile phase and detected at 365 nm.And fingerprint analysis of flavone glycosides in Flos Farfarae was performed by HPLC on ODS-BP C18 column(200 mm×4.6 mm,5 μm),eluted with acetonitrile-0.1% H3PO4 gradiently,and detected at 365 nm.Results The contents of quercetin and kaempferide in Flos Farfarae ralated to the habitats and processing obviously,but unrelated to growth way.The similarity of fingerprints of Flos Farfarae from different habitats and batches was higher.Conclusion The quantification method of quercetin and kaempferide in Flos Farfarae is convenient,accurate,stable,and could be used for quality control of flavone glycosides in Flos Farfarae.The contents of quercetin and kaempferide are obviously different from various Flos Farfarae.The method of the fingerprint analysis is feasible and reproductive,and could be used as the foundation for study on the fingerprints of flavone glycosides in Flos Farfarae.
