[摘要]
目的研究红芪Hedysarum polybotrys的化学成分和生物活性。方法采用硅胶柱色谱,Sephadex LH-20分离化学成分,根据波谱数据和理化性质确定化学结构;MTT法测定化合物对人肝癌细胞系HepG2的体外抑制作用,用半数抑制浓度(IC50)评价其抗肿瘤活性。结果共分离鉴定了11个化合物,分别为正二十四烷酸(Ⅰ)、正二十六烷酸(Ⅱ)、三油酸甘油酯(Ⅲ)、单棕榈酸甘油酯(Ⅳ)、3-甲氧基-4-羟基-反式苯丙烯酸正十六醇酯(阿魏酸正十六醇酯,Ⅴ)、丁香脂素(Ⅵ)、7-羟基-4′-甲氧基异黄酮(Ⅶ)、异甘草素(Ⅷ)、3-羟基-9-甲氧基紫檀烷(Ⅸ)、β-谷甾醇(Ⅹ)、胡萝卜苷(Ⅺ)。化合物Ⅸ对HepG2的IC50为10.69μmol/L。结论化合物Ⅰ~Ⅳ、Ⅵ为首次从该植物中得到;首次研究化合物Ⅸ的抗肿瘤活性,结果显示其对HepG2具有一定的细胞毒活性。

[Abstract]
Objective To investigate the chemcial constituents in Hedysarum polybotrys and their antitumor activities.Methods The chemical constituents were isolated by various column chromatographic methods,and their structures were elucidated mainly by NMR and MS evidences;Cytotoxicities of the purified compounds against human cancer cell lines HepG2 were evaluated by MTT method.Results Eleven compounds were isolated and identified as: n-tetracosanoic acid(Ⅰ),n-hexacosanic acid(Ⅱ),trioleic glyceride(Ⅲ),glycerol monopalmitate(Ⅳ),cetyl ferulate(Ⅴ),(+)syringaresinol(Ⅵ),7-hydroxy-4′-methoxyisoflavane(Ⅶ),isoliquiritigenin(Ⅷ),3-hydroxy-9-methoxypterocarpan(Ⅸ), β-sitosterol(Ⅹ),and daucosterol(Ⅺ).Conclusion Compounds Ⅰ—Ⅳ and Ⅵ are isolated from this plant for the first time.Compound Ⅸ shows the inhibitory activity on HepG2 with IC50 values of 10.69 μmol/L.
