[摘要]
目的 应用随机扩增多态性DNA(random amplified polymorphic DNA,RAPD)标记技术对广东产5个类群的栽培巴戟天进行遗传多样性研究。方法 应用15种引物对64个个体进行检测,应用Popgen32软件分析群体的遗传多样性。结果 共得到224条带,其中多态位点112个,多态百分率为50.00%。种群内多态位点百分率在37.05%~53.13%,平均为45.86%。Shannon指数和Nei指数均反映出巴戟天各类群遗传多样性有一定的变化。Shannon指数为0.287 6,各类群的Shannon指数在0.103 3~0.236 2,Nei指数为0.175 6,各类群的Nei指数在0.074 5~0.154 0。结论 目前栽培巴戟天居群间有一定的分化,并产生了多种农家类型,其中小叶巴戟天(POP2)遗传多样性明显高于其他类型,这些可能是导致目前栽培巴戟天质量产生变异的主要原因。

Objective ?Toinvestigatethegenetic diversityof fivepopulationsof cultivated Morinda?officinalis inGuangdongProvinceusingrandom amplified polymorphicDNA(RAPD).Methods ?Sixty-?four individuals?in?five populations?of cultivated M.officinalis were analyzed by RAPD markers?to?determine thegeneticvariationsamongthepopulations.Thedata ofgenetic diversitywereanalyzedwith?Popgen32software.Results ?The levels ofgeneticvariation and patterns of population?structure in M.?offi'cinalis were?investigated using RAPD markers.Of the?100 primers screened,15 primers produced?highly reproducible RAPDbands,usingtheseprimers,224discernible DNAfragmentsweregenerated?with?1 12(50.00%)polymorphic fragments,indicatingconsiderablegeneticvariation at the species level.?In contrast,there were relatively high levels ofpolymorphism at the populationlevel with thepercentage?ofpolymorphicbandsrangingfrom37.05%to53.1 3%,and the meanpercentageofpolymorphic loic(P-?45.86%).Genetic variationamong populationswas detected based on?Nei’S?genetic diversity analysis?(0.175 6),Shannon’S diversity index(0.287 6)for every population with Shannon?7S index 0.103 3-?0.236 2 and Nei?7S indexes 0.0745-O.1540.Conclusion ?There iS?a littlegeneticdifferentiationamong?populationsof cultivated M.officinalis and result in different cultivated types,the genetic diversity with?M.officinalis(POP2)is higherthan that of other populations.It maybe the main reason of the difference?of cultivated M.officinalis quality.?
