[摘要]
目的 观察血竭提取物对ApoE基因敲除(ApoE-/-)小鼠主动脉粥样硬化斑块成分、血脂及清道夫受体CD36 mRNA表达的影响,探讨血竭提取物稳定斑块的作用及可能机制。方法 33只6~8周龄ApoE-/-小鼠予高脂饮食喂养13周后,随机分为3组:模型组、血竭提取物(450.5 mg/kg)组、辛伐他汀(9.01 mg/kg)组(阳性对照组),每组11只。继续高脂喂养并给予相应药物治疗13周后,检测血脂,取主动脉根部4个切面,分别行HE染色和Movat染色,采用易损指数[(细胞外脂质成分+泡沫细胞)/(平滑肌细胞+胶原成分)]综合评价药物对小鼠主动脉斑块稳定性的影响。实时荧光定量PCR法检测主动脉内CD36 mRNA的表达。结果 给药13周后,血竭提取物组和辛伐他汀组的斑块易损指数较模型组均有不同程度的降低(P<0.01),两组间比较无显著差异(P>0.05)。血竭提取物组血清总胆固醇(TC)和甘油三酯(TG)水平与模型组比较显著降低(P<0.05、0.01),而主动脉内CD36 mRNA的表达与模型组比较亦显著降低(P<0.01)。结论 血竭提取物可通过改善斑块内部成分来稳定易损斑块,其机制与调节血脂、抑制清道夫受体CD36 mRNA的表达有关。

Objective ?To observe?the?effects?of dragon?7?S blood?extract?on?atherosclerotic plaque?content,blood lipids,andmRNAexpressionofscavenger receptorCD36 in aorta ofApoE-geneknockout?(ApoE-/-)mice and to explore the effect of this medicine on plaque stability and its possible mechanism.?Methods ?Thirty-three ApoE。/。mice?at 6-8 week-old were fed with a high-fat diet for 13 weeks and they?were divided randomly into three groups:model group,dragon?7?S blood extract(450.5 mg/kg)group,?Simvastatin(9.01 mg/kg)group(positive contr01),11?per group.Thereafter,thetWOdrug-treated?groupswere?igadministrated with relevant drug for another?1 3 weeks accompanied by feeding high-fat?diet.Then?all?the mice were?sacrificed?at?the end of?the experiment.Their blood was collected?for?determiningthe concentration of bloodlipids.The morphologyandcompositionof atheroscleroticplaques?in aortic roots were examined in four tissue sections.Four sections were chosen and stained with HE and?Movatstains,respectively,and?the?effect?of severaldrugs?on?plaque?stability?of ApoE-7‘mice was?evaluated by usingthe vulnerability index[(extracellular lipids+foam cells)/(smooth muscle cells+?collagens)].ThemRNA?expression?of CD36?in?aorta?of ApoE手micewas determined?byReal-time?fluorescent?quantitative PCR?technology.?Results ?After?treated?with?drugs?for?1 3?weeks。the?vulnerability indexes ofplaqueindragon?7?S blood?extractgroupand Simvastatingroupwere reduced in a?differentdegree(P<0.01),there was nosignificantdifference between the tWOgroups(P>0.05),and?the concentration of total cholesterol(TC)and triglyceride(TG)inserum of mice?indragon?7?S blood?extractgroupwassignificantly decreased(PConclusion ?Dragon?7?S blood?extract may?stabilize atheroscleroticplaquein aorta ofApoE‘/-mice by improving plaque contents,whose mechanism?may be related?tO the modulation of blood?lipids and inhibition of the mRNA expressions of scavenger?receptor CD36.
