[摘要]
目的 分析鉴定细脚拟青霉多糖PTPS-1的理化性质和结构特征,并观察其抗肿瘤活性。方法 采用DEAE-纤维素及Sephadex G-100柱色谱分离纯化得到PTPS-1;利用HPLC、GC、IR、部分酸水解及NMR分析PTPS-1的相对分子质量、组成和结构特征;采用CCK-8检测观察PTPS-1对小鼠脾细胞和PTPS-1刺激培养的小鼠腹腔巨噬细胞上清对Sp2/0细胞的增殖活性。结果 PTPS-1的纯度为98.977%,平均相对分子质量为1.84×104,由甘露糖和半乳糖组成,二者摩尔比为1.3∶1,糖链结构中主链含有α-(1→6)、α-(1→2)和β-(1→3)连接的糖苷键,支链含有Gal-(1→3)连接。PTPS-1可明显促进小鼠脾细胞的增殖(P<0.01),PTPS-1刺激培养的小鼠腹腔巨噬细胞上清能抑制Sp2/0细胞的增殖(P<0.01)。结论 PTPS-1是从细脚拟青霉中分离出的具有免疫调节和抗肿瘤活性的中性杂多糖。

Objective ?To investigatethephysicochemical properties,structuralcharacters and antitumor activity of a polysaccharide PTPS-1 from Paecilomyces tenutpes.Methods ?PTPS-1 was obtained by?DEAE-cellulose andSephadexG-100chromatographycolumn.The Methods ofHPLC,GC,IR,and NMR?were used?tOinvestigate the molecular weight,monosaccharide composition,andstructural characters of?PTPS-1.The CCK-8 kit was used?to determine the proliferationof PTPS-1 on murine splenocyte and the?supernatant of the murinemacrophagewith PTPS-1-stimulated onSp2/Ocells.Results ?PTPS-1,whose?molecularweight was 1.84×104 and puritywas 98.977%,contained mannose(Man)and galactogen?(Gal)as monosaccharide constitute in molar ratio of 1.3:1.It consisted of a backbone of a-(1-’6),a-(1?-2),andp(1-3)linkage,whosebranches were made?by Gal-(1·3)linkage.PTPS-1 significantly?stimulated the murine splenocyte proliferation and the supernatant of macrophageinhibited thegrowthof?Sp2/0cells?in vitro.Conclusion ?PTPS-1?is?a polysaccharide fromP.tenuipeswith immunomodulatory?and antitumoractivity.
