目的 研究半边旗二萜类化合物5F(11α-羟基-15-氧-16-烯-对映贝克杉烷-19酸)对人高转移卵巢癌细胞系HO-8910PM细胞内核转录因子κBP65亚基[NF-κB(P65)]、Smad3蛋白及NF-κB(P65)、ETS-1mRNA表达的影响,探讨其抗肿瘤的作用机制.方法 MTT法检测5F对HO-8910PM细胞增殖的影响;Westernblot分析NF-κB(P65)及Smad3蛋白的表达;RT-PCR分析NF-κB(P65)及ETS-1mRNA的表达.结果 5F能抑制HO-8910PM细胞的增殖,抑制率随剂量的增加而增加,具有剂量依赖性;25~100μmol/L5F作用HO-8910PM细胞24h后,NF-κB(P65)、Smad3蛋白表达水平明显下降,并呈剂量效应关系;5F明显下调NF-κB(P65)mRNA的表达,轻微下调ETS-1mRNA的表达.结论 5F抗肿瘤活性与NF-κB(P65)、Smad3蛋白和NF-κB(P65)、ETS-1mRNA的表达有关.

[Abstract]
Objective To study the effect of a diterpenoid 5F(ent-lla-hydroxy-15-OXO-kaur-16-an-19-oic acid)isoiated from Pteris semipinnata on the expressions of NF-KB(P65),Smad3 protein and NF-KB(P65),ETS-1 mRNA,and to investigate the antitumor mechanisms of 5FMethods MTT assay was used to examine the effect of 5F on proliferation of HO-8910PM cells.The expressions levels of NF-IcB(P65)and Smad3 proteins were assessed by Western blot analysis.RT-PCR assay was used to assess the expression levels of NF-KB(P65)and ETS-1 mRNAResults The proliferation of HO-8910PM cells can be inhibited by 5F in a dose-dependent manner.The expression levels of NF-xB(P65)and Smad3 proteins decreased obviously in HO-8910PM cells after treatment with 25-100 gmol/L 5F for 24 h.and the effects appeared in a dose-dependent manner.5F down-regulated significantly the expression of NF-KB(P65)mRNA.down-regulated gently the expression of ETS-1 mRNAConclusion The antitumor activity of 5F is involved in the expression of NF-KB(P65),Smad3 protein and NF-KB(P65),ETS-1 mRNA.
