[摘要]
目的 考察影响二苯乙烯苷(THSG)溶液稳定性的因素,并计算室温下其晶体粉末的t0.9值.方法 采用HPLC法和HPLC-MS联用法.结果 以Lichrospher 5-C18(250 mm×4.6 mm,5μm)为固定相,甲醇-水(4:6)为流动相,检测波长为310 nm.THSG进样量在0.14~2.80μg与峰面积线性关系良好,自制THSG纯度高达99.94%,pH 5.8时,THSG与水分子发生加成反应.强酸促使THSG水解为葡萄糖和苷元,并进一步降解为酚类化合物.强碱性条件下,THSG氧化为醌类化合物.在中性及弱碱性水溶液中较稳定,30 d内含量无明显变化.THSG晶体粉末室温下放置240 d含量无明显变化,t0.9值为1 098 d.结论 二苯乙烯苷在一定条件下是稳定的。

Object To study the factorswhich influenced the stability of stilbene glucosidel(tetrahy-droxystilbene-glucoside,THSG) solution,investigate and calculate the value of to.9 of THSG crystalpowder at room temperature·Methods HPLC and HPLC-MS Methods were used.Results The best chromatographic conditionswere to employLichrospher 5-C18 (250 mm X4.6mm,5 μm)as the stable phase,methanohvater (4:6) as mobile phase,310 nm as the detective w avelength·W ithin the range from 0.14to 2.80 μg,there w as a good linearity·The content of self-prepared THSG was up to 99·94%.Stability test show ed that w hen pH w as 5.8,an additional reaction occu rred betty een TH SG and H2O·Strong acids precipitated the hydrolyzation of THSG into glucose and aglycone,which then degraded into phenol compound.W hen strong bases existed,THSG was easily oxidized into quinine compound.Whereas itwas stable in a neutralorweak basic condition,the contentof THSG had not changed in 30 d.So was the THSG's crystalpowder stored at room temperature for 240d.The value of t0.9 was 1 098 d.Conclusion THSG is stable in some certain conditions.
