[摘要]
目的 从生物物理化学的角度,探讨黄连及其不同炮制品药性的客观真实性.方法 利用微量热法,测定了大肠杆菌在黄连不同炮制品总生物碱作用下的生长热谱曲线,得到了相应的生物热动力学参数,结合本草文献报道,综合分析不同炮制方法对黄连药性的影响.结果 黄连不同炮制品总生物碱均能不同程度地抑制大肠杆菌的生长,偏温性药姜制黄连、酒制黄连、吴萸黄连使大肠杆菌指数生长期的生长速率常数显著减小,生长代谢过程中热量释放显著增加.偏寒性药生黄连、醋制黄连、胆汁黄连也使生长速率常数减少,却使热量释放略增加,两者存在较稳定的差异.结论 微量热法可作为刻划中药药性的有效的新方法.

Object To probe into the Objectivity and authenticity of four properties(cold,heat,warm,and cool) of raw Coptid is Rizama (RCR)and their preparations from biophysics and biochemistry.M etbods M icrocalo rin etry w as a sed to ob taro the m etabo lic grow th pow ertin a curve of bacteria affected by the total alkaloid in differentpreparationsof RCR and biothermokinetic parameters.Combined with the records of ancient herbal literatures,the influence of properties by differentpreparations to RCR were synthetically evaluated.Results A 11 the effective total alkaloid in differentpreparations of RCR can restrain the Escherichia coli growth andmetabolign in different level.Drugs,such as CoptidisRhizana S tir-fried with g finger ju ice SCR GJ),Cop tid is Rhizamstir-fried with wfine CRW),and Cop tid is RhizaMstir-fried with Fructus Evod iae (CR FE),w ith a little WaProperty can decrease themultiplication rate constants of E·coli index and increase heat ou中ut in grow th m etablign rem arkably,w bile the drugs,such as RCR,Cop tid is Rhiza stirfried with vinegar (CRV),and Coptid is Rhizam stirfriedw ith gallbladder juice(CRGBJ),with a little Cold Property can decrease themultiplication rate constants and increase heat output a little.There w as a stable difference bettyeen them·Conclusion Microcalorinetry is a new and useful can for the study of the properties of tradition Chinese medicine.
