[摘要]
目的 评价菊花倍半萜烯内酯类化合物 (SLs)对鼻咽癌的抗肿瘤作用。方法 采用 SLs的活性成分parthenolide (PN)为受试物作用于人鼻咽癌高分化细胞株 CNE1,检测细胞毒性和诱导性凋亡指标,进行时间 -效应和剂量 -效应作用观察。结果 10~ 10 0 μm ol/ L 的 PN作用 6~ 36 h后与阴性对照组比较,CNE1细胞乳酸脱氢酶 (L DH)漏出率明显增加、细胞计数减少、细胞增殖抑制率升高 (P<0 .0 5 );细胞出现凋亡特征形态学改变,TU NEL阳性细胞、Sub G1亚群细胞百分数增加,细胞失贴壁率明显增高 (P<0 .0 5 );上述指标与 PN剂量存在明显相关 (P<0 .0 5 )和时间循序变化。结论 PN可通过对人鼻咽癌细胞毒性作用和诱导细胞凋亡发挥其抗肿瘤作用,提示 PN具有化学预防意义.

[Abstract]
Object To investigate the potential antitumor effects of sesquiterpene lactones (SLs) in the plants of chrysanthemum on human nasopharyngeal carcinoma (NPC). Methods Parthenolide (PN), the active component of SLs, was used as tested substance and exposed to NPC cell line CNE1. Cell toxicity and apoptosis indexes were determined at time effect and dose effect study. Results Compared with negative control, there were significantly increasing of lactate dehydrogenas LDH leakage and cell proliferation inhibition ratio as well as remarkable decrease in cell number, respectively in PN groups treated with 10 100 μmol/L PN for 6 36 h (P<0 05). There were high levels of TUNEL positive cell and SubG1 of cell cycle percentage, and significant increase in cell detachment ratio (P<0 05), while coupled with the characteristic apoptotic morphology changes in PN treated groups. The above indexes showed evident relative PN dose (P<0 05) and circulatory changes in time order. Conclusion It indicates that PN could induce cytotoxicity and apoptosis in exposed CNE1 cell, which correlated with its anticancer and chemoprevention effects on NPC.
