[摘要]
目的观察绞股蓝总皂苷(GP)对血管性痴呆大鼠大脑皮层及海马的DNA和RNA的影响。方法采用改良的Pulsinelli4-血管阻断(4-VO)方法建立大鼠血管性痴呆模型,用吖啶橙染色法进行GP对血管性痴呆大鼠大脑皮层及海马的DNA和RNA保护作用的研究。结果与正常对照组比较,血管性痴呆大鼠大脑皮层及海马的DNA和RNA吖啶橙染色后的荧光强度(反映DNA和RNA含量)明显减弱,GP200mg/kgig给药组大脑皮层及海马的DNA和RNA吖啶橙染色后的荧光强度强于血管性痴呆模型组,与正常对照组相似。结论GP可明显减轻血管性痴呆大鼠大脑皮层及海马的DNA和RNA损伤。

Object To o bserve the effect of gy penosides(GP) on the DNA and RNA in cerebralcortex and hippocampusu vascular dementia rat.Methods Modified Pulsinelli 4-vessel occlusion(4-VO) method was used to establish the models of vascular dementiarat.The acridine orange(AO) method was applied to study the protective effect of GP on the DNA and RNA of cerebral cortex and hippocampus in vascular dementiarat.Results The fluorescent intensity from the DNA and RNA in cerebral cortex and hippocampus of vascular dementia rat is abated as compared with the normal control group.The fluorescent intensity from DNA and RNA of the treatment group administrated GP(200 mg /kg,ig)significantly intensified,as compared with that of no n-treatment group,and is similar to that of normal group.Conclusion The injury to the DNA and RNA in cerebral cortex and hippocampus in vascular dementiarat was obviously decreased by GP.
