[摘要]
目的 应用神经行为畸胎学方法 评价豆腐果苷对大鼠仔代的神经行为毒性。方法 妊娠6~15d大鼠采用豆腐果苷0,27,270,1350mg/kg·d剂量连续ig染毒,17只孕鼠自然分娩。观察母鼠妊娠期和分娩后的生理、生育指标;160只仔鼠出生后的体重增长、早期生理发育和神经行为发育指标;随机选择32只仔鼠于10周龄时进行自动化操作行为测试。此外,对10周龄仔鼠24只进行脑组织的光镜观察和单胺类神经递质(去甲肾上腺素、多巴胺、5-羟色胺)的测定。整个实验采用双盲法。结果 未观察到豆腐果苷对大鼠的母体毒性;3个剂量组仔鼠的体重增长、早期生理发育及神经行为发育与对照组相比无显著性差异(P>0.05);操作行为测试中,仔鼠学习记忆能力无性别差异,3个剂量组仔鼠的学习成绩与对照组相比均无显著性差异(P>0.05);与对照组相比,各剂量组仔鼠脑组织均未观察到明显的形态学改变,脑神经递质含量也未见异常(P>0.05)。结论 未观察到胚胎期服用豆腐果苷对大鼠仔代神经系统的早期发育、神经行为功能和脑组织学的影响。

[Abstract]
Object To evaluat eneu robeh avioral effects of helicid on the off spring of rats received the heliciddu ring gestation with the methods of neurot oxicological teratology. Methods .The Wistar rats being pregnant for 6~15 dreceived helicid at 0,27,270 and 1350mg/kg·d byig. The physiological grow thindex of 17 rats were obs erved during pregnancy and after parturition.The weight-gaining,early physilolgical and neurogenicalde veloping index of 160 off spring was measured. The automaticoperation be haviortest for 32 off spring were carried out at the age of ten weeks. Moreover, the histological sections of off sprin g's brains were observedunder lightmicroscope and the level of monamineneurotrans mitterin brain,such as NA,DA,and 5-HT,was detected from 24 in fants of ten week sold. All the experiments were carried out indouble-blind manner.Results The neuro behavioraltoxicity was notob servedinall treatedrats.Compared with the control group,there were no signifi cant differences in the weight gaining, physiological and neuro behavioral development of off spring in three treated groups(P> 0.05);in operation behavior there were also no significant differences in all treated off spring groups(P> 0.05).N ogeneral histomo rphological alterations in brains of off spring were observed in all groups. The level of monamine neurotrans mitter in off spring's brains of all treated groups did n't change as compared with that of control group(P>0.05).Conclusion The administration of helicid to pregnant rats did not affect the early development of nervous syste m, neurobehavioral function and brain histology of off spring.
