目的 研究三七总皂苷 (panax notoginseng saponins,PNS)对烫伤早期大鼠心肌 Gsα-m RNA表达的影响。方法 通过 30 %体表面积皮肤度烫伤大鼠模型,应用 dot blotting杂交、心肌组织原位杂交、放射免疫分析等技术观察三七总皂苷对严重体表烫伤大鼠心肌 Gsα-m RNA表达水平、c AMP产生量、腺苷酸环化酶 (adenylylcyclase,AC)活性的影响。结果 大鼠心肌组织中 Gsα-m RNA表达量、cAMP含量、AC活性在烫伤后 3h均明显降低。 100,200 m g/ kg PNS明显增加烫伤大鼠心肌组织 Gsα-m RNA的表达,增加量与 PNS剂量呈显著正相关(r=0 .95,P<0 .0 5)。 100,200 mg/ kg使烫伤大鼠心肌组织 c AMP含量分别增加 2 1%,36 %,AC活性分别增强40 %,6 3% (P<0 .0 1)。结论 PNS可增加烫伤大鼠心肌 Gsα-m RNA表达,增强 AC活性,增加 c AMP生成.

[Abstract]
Object To investigate the effects of Panax notoginseng saponins (PNS) on myocardial Gsα-mRNA expression of seriously scalded rat. Methods A 30% skin full thickness scald model was produced by immersing rat in 95 ℃ water for 10 s. The effects of PNS on myocardial Gsα-mRNA level were observed with dot blotting hybridization and in situ hybridization technique; effects on cAMP and adenyl cyclase (AC) activities were determined with radioimmunoassay. Results Myocardial Gsα-mRNA, AC activity and cAMP content were reduced significantly 3h after scalding. PNS (100, 200 mg/kg) could markedly increase the level of myocardial Gsα-mRNA expression. The elevated quantity was correlated markedly with PNS dosage (r=0.95,P<0.05). In situ hybridization showed Gsα-mRNA expression was also increased. AC activity was enhanced by 40% and 63% (P<0.01) at 100, 200 mg/kg PNS respectively; cAMP content was increased by 21% and 36%, respectively. Conclusion PNS could improve myocardial Gsα-mRNA expression, enhance AC activity and increase cAMP content of seriously scalded rats.
