从新生牛主动脉分离获得单个内皮细胞,培养于微孔滤膜上直至形成致密单层。通过灌流 hanks液或含5g/L 白蛋白的 hanks液后,测定经10-4 m ol/L 组胺溶液处理或经10-4 mol/L 组胺溶液加10-4g/m L 当归补血汤处理的液体滤过系数(Kf)、液体滤过流量(Jv)和蛋白质渗透压反射系数(σ)。结果表明内皮单层经组胺处理后,Kf 和 Jv降低,σ升高;而当归补血汤能够抑制因组胺造成的 Kf 和 Jv降低,以及 σ升高。说明组胺能够引起血管内皮单层通透性的增加,而当归补血汤能够减轻组胺造成的内皮单层通透性的增加;其作用机制需进一步研究。

Blood Replenishing Angelica Decoction [Danggui Buxue Tang(DGBXT)]is a well known TCM prescription composed of Radix Angelicae Sinensis and Radix Astragali with the actions of invigorating “Qi” and enriching blood Its action to curtail endothelial permeability induced by histamine was studied Endothelial cells isolated from the aorta ofneonatal calf were cultured on polycarbonate microporus filter membrane to develop a confluent endothelial monolayer After purfused with either plain Hank's balanced salt solution or that containing5g/L albumin, the monolayer was treated with10-4 mol/L histamine for30min either with or without preincubation for60min with10-4g/mL of DGBXT Fluid filtration coefficient(Kf), filtration volume(Jv)and osmotic reflective coefficient(σ)of protein were then measured The findings showed that DGBXT could curtail the lowering of Kf and Jv and elevation of σ induced by histamine, indicating that DGBXT could inhibit the action of histamine on endothelial permeability, but its mechanism of actionneeds further study.
