用发根农杆菌 Agrobacterium rhizogenes R¹⁶⁰¹,A⁴,ATCC¹⁵⁸³⁴菌株感染菘蓝 Isatis indigoticaFort.同源四倍体的子叶10d后,在子叶切口处均能陆续长出毛状根。转化根在不含激素的 MS固体培养基上快速生长并表现出典型的毛根性状,经冠瘿碱分析证明确已被转化。建立了毛状根培养系统并比较了各种外界因子生长的影响

Hairy root of autotetraploid Isatis indigotica Fortune were induced from cotyledon leaf explants after 10 d inoculation with Agrobacterium rhizogenes, strains R1601, A4 and ATCC15834.Transformed roots grew rapidly on solid MS medium without hormones and showed typical hairy root phenotype.Transformation was confirmed by opine analysis.A culture system of hairy root was established and different extrinsic factors affecting the growth of hairy root was investigated.
