目的:建立当归的HPLC指纹图谱,并对其化学成分进行分析。方法:采用高效液相色谱法,以甲醇-0.05%磷酸水溶液系统进行线性梯度洗脱,测定了来源于岷县的多批当归药材和市售当归饮片的指纹图谱,应用相似度分析和聚类分析进行分类;并采用液相色谱-质谱联用(HPLC-MS)方法对岷县当归药材中的化学成分进行定性分析。结果:在选定的色谱条件下,得到当归HPLC指纹图谱,标定出12个共有峰;根据相似度分析和聚类分析的结果,将当归药材和当归饮片各分为1类。HPLC-MS鉴定出阿魏酸,6,7-二羟基藁本内酯,洋川芎内酯I、H、A,阿魏酸松柏酯,E-藁本内酯,Z-藁本内酯,3-正丁烯基苯肽共9个化学成分。结论:应用本方法评价当归的质量是可行的。

[Abstract]
Objective: To establish the HPLC fingerprint of Angelicae Sinensis Radix and analyze the chemical components by HPLC-MS. Methods: The HPLC method was used with Accurasil-C18 column (250 mm × 4.6 mm, 5 μm), and methanol -0.05% phosphoric acid was used as mobile phase in a gradient elution. The HPLC fingerprint was established for the determination of several batches of Angelicae Sinensis Radix grown in Minxian and praeparata Angelicae Sinensis Radix sold in the market and the similarity, and cluster analysi were carried out. The chemical components of Angelicae Sinensis Radix grown in Minxian were analysed by HPLC-MS. Results: Under the selected spectrum condition, 12 public peaks were shown in the HPLC fingerprint. Angelicae Sinensis Radix was classified into two groups based on the result of similarity and cluster analysi including Angelicae Sinensis Radix grown in Minxian and praeparata Angelicae Sinensis Radix sold in the market. Ferulic acid, 6,7-dihydroxydihydroligustilide, senkyunolide I, senkyunolide H, senkyunolide A, coniferyl ferulate, E-ligustilide, Z-ligustilide, and 3-butylidenephthalide were identified in chromatograms based on their MS data. Conclusion: This method is reasonable and reliable to the quality control of Angelicae Sinensis Radix.
