[摘要]
目的 基于HPLC-MS/MS技术建立桔梗八味颗粒定量分析方法,结合化学模式识别和熵权-TOPSIS分析综合评价其质量。方法 采用HPLC-MS/MS法分析10批桔梗八味颗粒中甘草苷、甘草素、芦丁、异鼠李素、没食子酸、木犀草素、槲皮素和阿魏酸;利用微生信平台、SIMCA 14.1软件对测定结果数据进行聚类分析(CA)、主成分分析(PCA)和正交偏最小二乘判别分析(OPLS-DA),并以变量重要性投影(VIP)值>1.0为原则筛选质量差异成分;基于熵权-TOPSIS分析对10批桔梗八味颗粒的质量优劣进行排序。结果 CA和PCA将10批桔梗八味颗粒分为两大类:S2、S5、S9批聚为一类,S1、S3、S4、S6~S8、S10批聚为一类。OPLS-DA模式下筛选出异鼠李素、芦丁、甘草素、没食子酸、甘草苷5个质量差异标志物。熵权-TOPSIS相对贴近程度在0.039~0.955,表明不同批次桔梗八味颗粒质量差异明显,其中S2、S5、S9批次排名靠前。结论 所建立方法快速、简单,可用于桔梗八味颗粒质量的综合评价。

[Abstract]
Objective To establish quantitative methods of Jiegeng Bawei Granules by HPLC-MS/MS method, and to comprehensively evaluate its quality combined with chemical pattern recognition analysis and entropy weight-TOPSIS. Methods The contents of liquiritin, liquiritigenin, rutin, isorhamnetin, gallic acid, luteolin, quercetin, and ferulic acid in 10 batches of Jiegeng Bawei Granules were quantitatively analyzed by HPLC-MS/MS method. The contents data were analyzed by CA, PCA, and OPLS-DA using bio-informatics platform and SIMCA14.1 software, and the VIP value greater above 1.0 was used as the index to screen the differential components. Entropy weight-TOPSIS method was used to rank the advantages and disadvantages of Jiegeng Bawei Granules quality of 10 batches. Results According to the results of CA and PCA, 10 batches of samples could be clustered into two categories: S2、S5、S9 were clustered into one category, and S1、S3、S4、S6~S8、S10 were clustered into one category. Under the OPLS-DA model, 5 difference markers isorhamnetin, rutin, liquiritigenin, gallic acid, and liquiritin were selected. Entropy weight-TOPSIS Euclidean closeness ranges was from 0.039 to 0.955, indicating that the quality of Jiegeng Bawei Granules varies greatly among different batches, with S2, S5, and S9 batches ranking high. Conclusion The established methods is rapid and simple, and can be to comprehensively evaluate the quality of Jiegeng Bawei Granules.
