[摘要]
目的 建立玄参药材的超高效液相色谱(UPLC)指纹图谱,结合化学计量学分析对玄参药材质量进行综合评价。方法 建立玄参药材的UPLC指纹图谱,采用聚类分析(CA)、主成分分析(PCA)、正交偏小二乘判别分析(OPLS-DA)和接收者操作特征(ROC)曲线分析对玄参药材进行质量对比。结果 玄参UPLC指纹图谱分析符合方法学考察要求,CA和PCA分析结果相似,但PCA分析呈更强的地域性特征,OPLS-DA可以实现恩施、宜昌两地样本的有效区分,并筛选出哈巴苷、安格洛苷C在内的4个差异性成分,ROC分析表明了OPLS-DA模型具有较强的产地分类能力。结论 玄参药材UPLC指纹图谱结合化学计量学分析可以评价玄参药材的质量,为玄参药材的质量分析提供参考。

[Abstract]
Objective To establish UPLC fingerprint of Scrophulariae Radix and comprehensively evaluate quality of Scrophulariae Radix by chemometric analyses. Methods UPLC fingerprint of Scrophulariae Radix was constructed. Cluster analysis (CA), principal component analysis (PCA), orthogonal partial least squares-discriminant analysis (OPLS-DA) and receiver operating characteristic (ROC) curves were applied to evaluate the quality of Scrophulariae Radix. Results The UPLC fingerprint method for Scrophulariae Radix met the requirements of methodological validation. Results of CA and PCA were similar, whereas PCA displayed stronger geographical characteristics. OPLS-DA effectively distinguished samples from Enshi and Yichang area, and four differential components including harpagoside and angoroside C were screened out. ROC analysis indicated that the OPLS-DA model possessed strong classification ability for origins. Conclusion UPLC fingerprint of Scrophulariae Radix combined with chemometric analyses can comprehensively evaluate Scrophulariae Radix, providing a reference for quality analysis of Scrophulariae Radix.
