[摘要]
目的 建立HPLC-水凝粒子激光计数检测器(NQAD)法测定芪冬颐心颗粒中黄芪甲苷、麦冬皂苷D、丹参酮ⅡA、人参皂苷Rb1、茯苓酸、梓醇、桂皮醛、淫羊藿苷、绿原酸和柚皮苷。方法 使用InfinityLab Prooshell 120 SB-C18色谱柱(150 mm×4.6 mm,4 μm);流动相:0.1%甲酸-乙腈,梯度洗脱;柱温:40 ℃;进样量:10 µL;体积流量:0.9 mL/min;NQAD参数为蒸发温度:90 ℃;喷雾温度:30 ℃;倍率:1;出口体积流量:1.4 mL/min;平滑度:5 s;线性补正:开。结果 黄芪甲苷、麦冬皂苷D、丹参酮ⅡA、人参皂苷Rb1、茯苓酸、梓醇、桂皮醛、淫羊藿苷、绿原酸和柚皮苷的平均回收率分别为98.88%、97.81%、97.23%、96.79%、98.11%、97.21%、98.08%、97.98%、97.38%、97.72%,RSD值分别为0.86%、1.14%、1.27%、1.32%、1.05%、1.42%、0.99%、1.11%、1.17%、1.47%。结论 方法高效、快速、准确,可以应用于芪冬颐心颗粒的质量控制。

[Abstract]
Objective To develop an HPLC-NQAD method for determination of astragaloside, ophiopogonin D, tanshinone ⅡA, ginsenoside Rb1, pachymic acid, catalpol, cinnamaldehyde, icariin, chlorogenic acid, and naringin in Qidong Yixin Granules. Methods The chromatographic separation was performed on InfinityLab Prooshell 120 SB-C18 column (150 mm × 4.6 mm, 4 μm) with a gradient elution of 0.1% formic acid-acetonitrile. Column temperature was 40 ℃, injection volume was 10 µL, and volumetric flow rate was 0.9 mL/min. The NQAD parameter was as following: evaporation temperature: 90 ℃, spray temperature: 30 ℃, magnification: 1, export volume flow rate: 1.4 mL/min, smoothness: 5 s, and linear correction: on. Results The average recoveries of astragaloside, ophiopogonin D, tanshinone ⅡA, ginsenoside Rb1, pachymic acid, catalpol, cinnamaldehyde, icariin, chlorogenic acid and naringin were 98.88%, 97.81%, 97.23%, 96.79%, 98.11%, 97.21%, 98.08%, 97.98%, 97.38%, and 97.72%, respectively, with RSD values of 0.86%, 1.14%, 1.27%, 1.32%, 1.05%, 1.42%, 0.99%, 1.11%, 1.17%, and 1.47%, respectively. Conclusion The method is efficient, fast, and accurate, and can be applied to the quality control of Qidong Yixin Granules.
