[摘要]
目的 采用超高效液相色谱-质谱(UPLC-QTOF/MS)鉴定蕨麻中化学成分。方法 Acquity UPLC HSS T3色谱柱(100 mm×2.1 mm,1.8μm);流动相:0.05%甲酸水溶液–乙腈,梯度洗脱;体积流量:0.3 mL/min;柱温:40 ℃;进样体积:2 μL。ESI电喷雾离子源正负离子扫描,二级IDA扫描模式下采集碎片数据。通过Sciex OS 2.0数据处理软件结合数据库、对照品、部分文献和天然产物裂解规律对蕨麻水提液中化学成分进行鉴定。结果 共鉴定出53种化学成分,其中氨基酸类10个,酚酸类8个,萜酸类4个,黄酮类7个,季胺类生物碱1个,其他类23个。以野鸦椿酸、鞣花酸为例进行碎片离子裂解推测。结论 该方法准确稳定,适用于蕨麻中化学成分的鉴定,为后续的质量控制和物质基础研究提供参考。

[Abstract]
Objective To identify the chemical constituents in Potentillae Radix by UPLC-QTOF/MS. Methods The separation was carried out on Acquity UPLC HSS T3 column (100 mm × 2.1 mm, 1.8 μm) with the mobile phase of 0.05% formic acid - acetonitrile by gradient elution. The flow rate was 0.3 mL/min, column temperature was 40 ℃, and the volume of sample injection was 2 μL. Electrospray ionization (ESI) was used. Positive and negative ion scanning of ESI electric spray ion source was used, and fragmented data were collected under the second level IDA scanning mode. Identification of chemical components in Potentillae Radix water extract were speculated using Sciex OS 2.0 data processing software combined with databases, reference substances, some literature, and natural product cleavage pathways. Results A total of 53 chemical constituents were identified, including 10 amino acids, 8 phenolic acids, 4 terpene acids, 7 flavonoids, 1 quaternary amine alkaloid, and 23 others. The fragmentation process of fragment ions was speculated taking gallic acid and tannic acid as example. Conclusion The method is accurate, stable, and suitable for identification of chemical constituents in Potentillae Radix, and provides reference for subsequent quality control and material basis research.
