[摘要]
目的 探讨芩百清肺浓缩丸对肺炎支原体肺炎小鼠肺上皮间质转化的影响。方法 将Balb/c小鼠随机分为对照组、模型组、阿奇霉素(0.03 g/kg)组及芩百清肺浓缩丸高、中、低剂量(4.6、2.3、1.15 g/kg)组,每组10只。除对照组外,其余各组小鼠进行支原体滴鼻,每只滴入20 μL 1×107 CCU肺炎支原体菌液,建立肺炎支原体感染模型。支原体肺炎模型建立成功后各组分别ig给予小鼠相应药物,连续ig 14 d。实验结束后取出小鼠肺组织,显微镜下观察各组小鼠肺组织苏木精–伊红(HE)染色、Masson染色以及免疫组化的肺组织变化。RT-PCR法检测各组小鼠肺组织E-钙黏蛋白(E-cadherin)、转化生长因子-β1(TGF-β1)和波形蛋白(Vimentin)mRNA表达量。结果 与模型组相比,芩百清肺浓缩丸组小鼠肺组织结构明显得到改善,结构清晰,组织形态规整。与模型组比较,芩百清肺浓缩丸组小鼠肺组织E-cadherin mRNA及蛋白表达增高;TGF-β1、Vimentin mRNA及蛋白表达显著降低(P<0.05)。结论 芩百清肺浓缩丸能够降低肺组织中TGF-β1、Vimentin的表达,增加E-cadherin的表达,从而抑制上皮间质转化。

[Abstract]
Objective To investigate the effect of Qinbai Qingfei Concentrated Pills on pulmonary epithelial interstitial transformation in mice with mycoplasma pneumoniae pneumonia. Methods Balb/c mice were randomly divided into control group, model group, azithromycin (0.03 g/kg) group and Qinbai Qingfei Concentrated Pills high, medium, and low-dose (4.6, 2.3, 1.15 g/kg) groups, with 10 mice in each group. Except the control group, mice in other groups received mycoplasma nasal drops, each of which was injected with 20 μL 1× 107 CCU mycoplasma pneumoniae solution to establish mycoplasma pneumoniae infection model. After the successful establishment of mycoplasma pneumonia model, mice in each group were given corresponding drugs intragastrically and intragastrically for 14 d. At the end of the experiment, the lung tissues of mice were taken out, and the changes of the lung tissues with hematoxylin-eosin (HE) staining and Masson staining were observed under the microscope. After the experiment, the lung tissues of mice were taken out, and the changes of hematoxylin-eosin (HE) staining, Masson staining, and immunohistochemical lung tissue were observed under the microscope. The mRNA expressions of E-cadherin, TGF-β1, and Vimentin in lung tissues of mice in each group were detected by Real-time PCR. Results Compared with model group, the lung tissue structure of mice in Qinbai Qingfei Concentrated Pills group was obviously improved, with clear structure and regular morphology. Compared with model group, mRNA and protein expression of E-cadherin in lung tissue of mice in Qinbai Qingfei Concentrated Pills group were increased. The mRNA and protein expressions of TGF-β1 and Vimentin were significantly decreased (P < 0.05). Qinbaiqingfei concentrated pills can decrease the expression of TGF-β1 and Vimentin, and increase the expression of E-cadherin in lung tissue, so as to inhibit the epithelial mesenchymal transformation.Conclusion Qinbai Qingfei Concentrated Pills can decrease the expression of TGF-β1 and Vimentin, and increase the expression of E-cadherin in lung tissue, so as to inhibit the epithelial mesenchymal transformation.
