[摘要]
目的 建立HPLC法同时测定乙肝宁颗粒中鸡矢藤次苷甲酯、车叶草苷酸、车叶草苷、丹参素、丹酚酸B、丹参酮ⅡA、毛蕊异黄酮葡萄糖苷和芒柄花素。方法 采用Kromasil C18色谱柱(250 mm×4.6 mm,5μm);流动相:乙腈-0.1%甲酸溶液,梯度洗脱;检测波长分别为238 nm(0~21.0 min检测鸡矢藤次苷甲酯、车叶草苷酸和车叶草苷)、280 nm(21.0~40.0 min检测丹参素、丹酚酸B和丹参酮ⅡA)和254 nm(40.0~55.0 min检测毛蕊异黄酮葡萄糖苷和芒柄花素);体积流量:1.0 mL/min;柱温:30℃;进样量为10 μL。结果 鸡矢藤次苷甲酯、车叶草苷酸、车叶草苷、丹参素、丹酚酸B、丹参酮ⅡA、毛蕊异黄酮葡萄糖苷和芒柄花素分别在1.49~37.25、0.76~19.00、1.07~26.75、0.99~24.75、12.61~315.25、2.43~60.75、0.69~17.25、1.86~46.50μg/mL线性关系良好(r ≥ 0.999 1);各成分平均加样回收率分别为98.44%、98.12%、99.36%、97.44%、100.00%、99.19%、96.98%、98.93%,RSD值分别为0.71%、1.47%、0.98%、1.35%、0.85%、1.13%、1.02%、1.29%。结论 该方法操作简便、重复性好,可用于乙肝宁颗粒中多指标性成分的质量控制。

[Abstract]
Objective To establish an HPLC method for simultaneous determination of feretoside, asperulosidic acid, asperuloside, danshensu, salvianolic acid B, tanshinone ⅡA, calycosin 7-O-β-D-glucopyranoside and formononetin in Yiganning Granules. Methods The chromatographic separation was carried out on Kromasil C18 column (250 mm×4.6 mm, 5 μm) with the mobile phase consisting of acetonitrile-0.1% formic acid solution with gradient elution at a flow rate of 1.0 mL/min.The column temperature was 30℃. The detection wavelength were set at 238 nm for feretoside, asperulosidic acid and asperuloside, 280 nm for danshensu, salvianolic acid B and tanshinone ⅡA, and 254 nm for calycosin 7-O-β-D-glucopyranoside and formononetin. Results Feretoside, asperulosidic acid, asperuloside, danshensu, salvianolic acid B, tanshinone ⅡA, calycosin 7-O-β-D-glucopyranoside and formononetin showed good linear relationships within the ranges of 1.49-37.25, 0.76-19.00, 1.07-26.75, 0.99-24.75, 12.61-315.25, 2.43-60.75, 0.69-17.25, 1.86-46.50 μg/mL (r ≥ 0.999 1), whose average recoveries were 98.44%, 98.12%, 99.36%, 97.44%, 100.00%, 99.19%, 96.98%, and 98.93% with the RSD values of 0.71%, 1.47%, 0.98%, 1.35%, 0.85%, 1.13%, 1.02%, and 1.29%, respectively. Conclusion The method is easy to operate and repeatable, can be used for quality control of multiple indicator components in Yiganning Granules.
