全球乳腺癌发病率呈逐年上升趋势,已经成为当前社会的重大公共卫生问题。细胞周期蛋白依赖性激酶(CDK)4/6抑制剂将细胞周期阻滞于G1期,从而起到抑制肿瘤增殖的作用。ribociclib属于高度特异性CDK4/6双重抑制剂,可靶向抑制D1/CDK4、D3/CDK6细胞周期,具有潜在的抗肿瘤活性。临床试验结果表明ribociclib可用于乳腺癌、黑色素瘤、非小细胞肺癌、畸胎瘤、脂肪肉瘤和胶质母细胞瘤的治疗。

The incidence of breast cancer is increasing year by year, and has become a major public health problem in the world. Cycle dependent protein kinase (CDK) 4/6 inhibitor can block cell cycle in G1 phase, which can inhibit tumor growth. Ribociclib is a highly specific protein kinases 4/6 inhibitor, which can inhibit D1/CDK4 and D3/CDK6 cell cycle, and has potential antitumor activity. The results of clinical trials indicate that ribociclib has clinical efficacy in treatment of breast cancer, melanoma, non-small cell lung cancer, teratoma, liposarcoma, and glioblastoma.
