[摘要]
目的 建立三七总皂苷中三七皂苷K的超高效液相色谱(UPLC)测定的方法。方法 采用Cosmosil Cholester色谱柱(50 mm×2.0 mm,2.5 μm);流动相:[水–乙腈(85:15)]–乙腈,进行梯度洗脱;体积流量0.5 mL/min;检测波长203 nm;柱温30 ℃;进样量2 μL。结果 三七皂苷K在2.020~101.000 μg/mL与峰面积呈良好的线性关系(r=0.999 9),平均回收率为99.50%,RSD值为1.35%。结论 建立的UPLC法操作简便、结果可靠、重复性好、专属性强,可用于三七总皂苷中三七皂苷K的质量控制。

[Abstract]
Objective To establish a method for determination of notoginsenoside K in Notoginseng total saponins by ultra performance liquid chromatography (UPLC) method. Methods The determination was carried out on Cosmosil Cholester column (50 mm×2.0 mm, 2.5 μm). The mobile phase consisted of [water-acetonitrile (85:15)]-acetonitrile with gradient elution at a flow rate of 0.5 mL/min. The detection wavelength was set 203 nm and the column temperature was at 30 ℃ with injection volume of 2 μL. Results Notoginsenoside K had good linearity in the ranges of 2.020-101.000 μg/mL (r=0.999 9). The average recovery was 99.50% with RSD of 1.35%. Conclusion The method has simple operation, accurate results, good reproducibility, and specificity which can be used in the quantity control of notoginsenoside K in Notoginseng total saponins effectively.
