目的 建立同时测定银屑优化颗粒中芍药苷、甘草苷、落新妇苷、迷迭香酸和甘草酸5种成分的HPLC-DAD测定方法。方法 采用高效液相色谱波长切换法。Lichrospher C18色谱柱(250 mm×4.6 mm,5 μm);流动相为乙腈–0.1%磷酸溶液,梯度洗脱;检测波长在230、254、290 nm切换;体积流量为1 mL/min;柱温为30 ℃;进样体积10 μL。结果 5种成分的线性关系良好。芍药苷在69.12~691.20 ng线性关系良好,平均加样回收率为97.45%,RSD值为1.54%;甘草苷在39.02~390.20 ng线性关系良好,平均加样回收率为97.10%,RSD值为1.90%;落新妇苷在30.03~300.30 ng线性关系良好,平均加样回收率为99.98%,RSD值为1.22%;迷迭香酸在7.82~78.20 ng线性关系良好,平均加样回收率为100.83%,RSD值为1.36%;甘草酸在31.09~310.90 ng线性关系良好,平均加样回收率为97.52%,RSD值为1.49%。结论 本方法具有方法简便、快速、准确等特点,可同时测定银屑优化颗粒中5种活性成分。

[Abstract]
Objective To establish a quantification method to determine paeoniflorin, liquiritin, astiblin, rosmarinic acid, and glycyrrhizic acid in Yinxie Youhua Granules by HPLC-DAD method. Methods The components were determined by HPLC simultaneously with changing ultraviolet-visible wavelength. The column was Lichrospher C18 (250 mm×4.6 mm,5 μm). The mobile phase was acetonitrile- 0.1% phosphoric acid solution in gradient elution mode. The detection wavelength was set at 230, 254 and 290 nm. The injection volume was 10 μL at the flow rate of 1.0 mL/min. The temperature of column was set at 30 ℃. Results The calibration curves of five active components were excellent. Paeoniflorin had a good linearity in the range of 69.12 — 691.20 μg and its average recovery was 97.45% with RSD of 1.54%. Liquiritin had a good linearity in the range of 39.02 — 390.20 μg and its average recovery was 97.10% with RSD of 1.90%. Astiblin had a good linearity in the range of 30.03 — 300.30 μg and its average recovery was 99.98% with RSD of 1.22%. Rosmarinic acid had a good linearity in the range of 7.82 — 78.20 μg and its average recovery was 100.83% with RSD of 1.36%. Glycyrrhizic acid had a good linearity in the range of 31.09 — 310.90 μg and its average recovery was 97.52% with RSD of 1.49%. Conclusion The established method is convenient and accurate with a satisfactory separation and high sensitivity. It can be used to determine the five components in Yinxie Youhua Granules with the same chromatogram condition.
