[摘要]
目的 研究生半夏对HepG2细胞增殖和转氨酶释放量的影响。方法 分别用石油醚、醋酸乙酯、95%乙醇、纯水提取生半夏,将提取物作用于HepG2细胞,设置对照组、5-氟尿嘧啶(100 mg/L)组和半夏提取物组(剂量分别为0.1、1、10、20 mg/L)。24 h后,MTT法测定生半夏对HepG2细胞增殖的影响;48 h后,测定培养上清中谷丙转氨酶(ALT)、天冬氨酸转氨酶(AST)的水平。结果 与对照组相比,生半夏各组分在剂量不高于10 mg/L时毒性很小,20 mg/L对细胞增殖有较强抑制作用,并显著性提高ALT释放量。不同组分的细胞毒性强弱趋势为石油醚>醋酸乙酯>95%乙醇>纯水。结论 生半夏对HepG2细胞有一定细胞毒性,表现为抑制细胞增殖和损伤细胞膜,其作用强度与剂量、提取溶剂有关。 关键词:生半夏;HepG2细胞;细胞增殖;转氨酶释放

[Abstract]
Objective To study the effect of Pinelliae Rhizoma on the cell proliferation and transaminase release of HepG2 cells. Methods Ligroin, acetidin, 95% ethanol, and water were used to extract the components from Pinelliae Rhizoma, and the extracts were added to HepG2 cells. Six groups such as normal (without drugs), control (5-fluorouracil, 100 mg/L), and Pinelliae Rhizoma extracts (0.1, 1, 10, and 20 mg/L) groups were set up. After 24 h, cell multiplication was determined by MTT assay. After 48 h, glutamicpyruvic transaminase (ALT) and aminotransferase (AST) in supernatant of culture medium were tested by kits. Results Compared with the control group, the toxicity of Pinelliae Rhizoma was weak when the dosages were no higher than 10 mg/L. When the dosage came to 20 mg/L, the toxicity was strong, and the release of ALT was enhanced significantly. The precedence ordering of toxicity of different extracts was ligroin > acetidin> 95% ethanol > water. Conclusion There are toxicity of Pinelliae Rhizoma on HepG2, and it is reflected by the inhibition of cell proliferation and cell membrane injury. The intensity is related with the processing methods and extracting solvent.
