[摘要]
目的 建立腺毛黑种草子中山柰酚的HPLC测定方法。方法 采用高效液相色谱法,色谱柱为Symmetry RP C18(150 mm×4.6 mm,5 μm)色谱柱,流动相为甲醇–0.4%磷酸水溶液(70∶30),体积流量1.0 mL/min,检测波长365 nm,进样量10 μL。结果 山柰酚在0.022 4~0.134 4 μg呈良好的线性关系(r=0.999 9),平均回收率为97.59%,RSD值为1.29%(n=9)。结论 该方法操作简便、准确、重复性好,可用于腺毛黑种草子中山柰酚的测定。

[Abstract]
Objective To establish an HPLC method for determination of kaempferol in the seeds of Nigella glandulifera. Methods HPLC was carried out on Symmetry C18 column (150 mm×4.6 mm, 5 μm) with methanol-0.4% phosphoric acid (70∶30) as mobile phase. The detection wavelength was 365 nm and injection volume was10 μL at the flow rate of 1.0 mL/min. Results The calibration curve was linear over the range of 0.022 4—0.134 4 μg (r=0.999 9) for kaempferol. The average recovery rate was 97.59% with RSD value at 1.29%. Conclusion The established method is rapid, sensitive, and accurate, and could be used for the determination of kaempferol in the seeds of N. glandulifera.
