目的 建立用反相高效液相色谱（RP-HPLC）法测定家兔眼房水中头孢匹胺钠的方法。 方法 将给予头孢匹胺钠后抽取的兔眼房水经去蛋白处理后，采用RP-HPLC分析。流动相为磷酸二氢钾溶液(0.05 mol/L)-甲醇（73∶27），检测波长254 nm，体积流量1.0 mL/min，柱温30 ℃。结果 头孢匹胺钠在0.12～2.50 μg/mL线性关系良好（r＝0.999 ），平均回收率为（94.30±0.96）％，RSD＜4％。结论 本法准确，重现性好，适用于眼房水中头孢匹胺钠的分析。
Objective To establish a RP-HPLC method to determine the Cefpiramide level in rabbit ocular aqueous.Methods The proteins in samples were removed by precipitation. The RPHPLC method was performed by using sodium dihydrogen phosphate (aqueous solution， 0.05 mol/L) and methanol in a ratio of 73∶27 as mobile phase at a flow rate of 1.0 mL/min. The wavelength of detection was 254 nm and columntemperature was at 30 ℃.Results The calibration curves were linear over the range of 0.12～2.5 μg/mL and with a correlation coefficient of 0.999 9. The average recovery was (94.30± 0.96)％ and the RSD less than 4%.Conclusion The HPLC method is accurate and reliable for the determination of ocular Cefpiramide in rabbit eyes.
