[摘要]
目的 初步探讨菊苣降尿酸的有效成分,并对其降尿酸机制、安全性进行初探。方法 迪法克鹌鹑50只(雄性),随机分为5组,即对照组、模型组、苯溴马隆组(20 mg/kg)、混合物大剂量组(菊苣酸、绿原酸、秦皮甲素剂量均为150 mg/kg)、混合物小剂量组(菊苣酸、绿原酸、秦皮甲素剂量均为50 mg/kg),每组10只。除对照组喂鹌鹑普通饲料外,其余各组均给予高嘌呤饲料(普通饲料拌入酵母提取物15 g/kg)复制高尿酸血症模型。给药3周,动态观察鹌鹑血清尿酸(UA)水平、肝功能指标丙氨酸转氨酶(ALT)和天冬氨酸转氨酶(AST)水平、肾功能指标肌酐(Cr)和尿素氮(BUN)水平、UA代谢相关酶黄嘌呤氧化酶(XOD)和腺苷脱氨酶(ADA)活性的变化。结果 造模期间,与对照组相比,第7~21天模型组鹌鹑血清UA水平显著升高(P<0.05);与模型比相比,第7~21天混合物大、小剂量组鹌鹑血清UA水平显著降低(P<0.05);混合物大、小剂量组鹌鹑血清的ALT、AST、Cr、BUN水平未显著升高(P>0.05),且各给药组鹌鹑血清XOD、ADA活性均显示不同程度的抑制。结论 菊苣降尿酸活性成分可能为绿原酸、秦皮甲素、菊苣酸,安全性较好,其发挥药效的机制可能与抑制XOD、ADA活性有关。

[Abstract]
Objective To investigate the active ingredients of chicory on uric acid-lowering, and make a preliminary study on the safety as well as mechanism of uric acid-lowering. Methods Fifty quails were evenly randomized into five groups, namely normal group, model group, benzbromarone (20 mg/kg) group, high-dose and low-dose mixture groups (150, and 50 mg/kg), 10 quails in each group. Except for the normal group, the quails in other groups were given high purine diet (ordinary forage mixed with 15 g/kg of yeast extract powder) to induce hyperuricemia model. And then we observed the changes of UA, ALT, AST, Cr, BUN, XOD, and ADA levels in serum during the treatment. Results During the molding period, model group of serum UA level significantly increased (P<0.05) compared with normal group in 7-21 d; Compared with the model group, the serum UA level of high-dose and low-dose mixture groups significantly decreased (P<0.05), ALT, AST, Cr and BUN levels did not significantly increase (P>0.05) in 7-21 d; The XOD and ADA levels showed different degrees of inhibition. Conclusion Chlorogenic acid, aesculin, as well as chicoric acid has the effect on lowering serum uric acid level in quail hyperuricemia model, which may be associated with reducing the activities of XOD and ADA levels.
