[摘要]
目的 考察大孔树脂分离复方黑骨藤药物有效部位的工艺条件。方法 以黑骨藤中杠柳苷、秦艽中龙胆苦苷和元胡中延胡索乙素为检测指标,采用HPLC法考察3种大孔树脂(D-101、AB-8、NKA-9)对杠柳苷、龙胆苦苷和延胡索乙素的吸附分离性能,并考察分离纯化杠柳苷、龙胆苦苷和延胡索乙素成分的最佳工艺条件。结果 用大孔树脂D-101作为填料,样品与填料的比为1:10,样品质量浓度为50 mg/mL,上样后吸附1 h,用60%乙醇为洗脱液,在pH 7.0条件下,20 BV洗脱1 h。结论 该优选的工艺简便可行,大孔树脂D-101可作为分离纯化复方黑骨藤药物中杠柳苷、龙胆苦苷和延胡索乙素成分的有效方法。

[Abstract]
Objective To investigate the technology for the separation of the active ingredients from Compound Periploca forrestil (CPF) by macroporous resin. Methods The absorption and separation properties of three kinds of absorption resins (D-101, AB-8, and NKA-9) were studied and the separation and purification technological process of glucoperiplocymarin, gentiamarin, and tetrahydropalmatine from CPF were optimized by HPLC with glucoperiplocymarin in P. forrestil, gentiamarin in Gentiana macrophylla, and tetrahydropalmatine in Corydalis yanhusuo as indexes. Results Using macroporous resin D-101 as filler, the rate of sample and macroporous adsorption resin was 1:10. The concentration of the sample was 50 mg/mL, adsorbing for 1 h and using 60% ethanol as eluent in the conditions of pH 7.0. The resin was washed by 20 BV in 1 h. Conclusion This technology is simple and feasible, and the process with macroporous resin D-101 is an effective method to separate and purify glucoperiplocymarin, gentiamarin, and tetrahydropalmatine from CPF.
